摘 要:目的:探讨丹参对豚鼠耳蜗缺血再灌注损伤(IRI)后细胞凋亡及FasL蛋白表达的影响。方法:健康豚鼠72只,随机分成正常组、假手术组、丹参干预假手术组、模型组和干预组(术前腹腔注射丹参注射液,每天1次共7 d),模型组和干预组再各分成3个亚组:缺血30 min组、再灌注6 h组及再灌注24 h组。用HE染色法观察缺血及再灌注不同时间段耳蜗各部位的形态学改变,用免疫组织化学法检测内耳FasL的表达,用原位凋亡法(TUNEL法)检测内耳细胞凋亡情况。结果:正常组、假手术组、丹参干预假手术组内耳罕见凋亡细胞,FasL为弱阳性表达;模型组Corti器、血管纹、螺旋神经节每个时间点均有细胞凋亡,FasL为阳性表达,与正常组比较有显著性差异(P<0.05);干预组Corti器、血管纹、螺旋神经节每个时间点均有细胞凋亡,FasL为阳性表达,程度较模型组减弱,与模型组比较有显著性差异(P<0.05)。结论:FasL参与了耳蜗IRI后的细胞凋亡,丹参可能通过抑制FasL蛋白的表达使凋亡细胞减少而对IRI后的耳蜗起保护作用。Objective:To investigate the effects of salvia miltiorrhiza on the cell apoptosis and the expression of apoptosis-assoeiated protein Fas ligand(FasL) in cochlear of guinea pigs after ischemia reperfusion injury. Methods:Total 72 healthy guinea pigs were divided into the normal group,sham operation group,sham opera-tion group treated with salvia miltiorrhiza,model group and intervention group treated with salvia miltiorrhiza everyday by intraperitoneal injection for 7 days before onset of ischemia. Model group and intervention group were divided into 3 subgroups:30 minutes after ischemia,6 hours after reperfusion and 24 hours after reperfusion. The histochemical features of cochlear were observed by HE staining. The expression of FasL protein in cochlear was studied by immunohistochemisty. The cell apoptosis was studied by TUNEL. Results: There were no cell apoptosis found in the normal group,the control group and sham operation group treated with salvia miltiorrhiza,with the weakly positive expression of FasL protein. Obvious cell apoptosis at differ- ent times presented in the organ of Corti ,stria vaseulars and spiral ganglion of model group,with the strong expression of FasL,which showed significant difference compared with those of normal group(P〈0.05). The number of apoptotic cells in intervention group were lesser than those of model groups (P〈0.05). Conclusion:Salvia miltiorrhiza could inhibit cell apoptosis by down-regulating the expression of FasL protein, which may be one of mechanisms of neuroprotective effects of salvia miltiorrhiza.
