摘  要:目的:建立测定戊己丸中4种生物碱类成分含量的方法。方法:采用反相高效液相色谱法,固定相为Diamonsil-C18柱,乙腈-0.05 mol·L-1磷酸二氢钾溶液(48∶52)(每100 mL中加入十二烷基硫酸钠0.4 g)为流动相,流速为1.0 mL·min-1,检测波长为284 nm,柱温为25℃。结果:盐酸小檗碱、盐酸表小檗碱、盐酸黄连碱、盐酸巴马汀分别在5.0~50.0,0.4~4.0,2.0~20.0,2.0~20.0 g·mL-1呈良好的线性关系;平均回收率分别为102.4%,100.1%,100.0%,102.0%;方法精密度RSD分别为0.36%,1.50%,1.60%,0.86%(n=6)。结论:该法可用于戊己丸的质量控制。Objective: To establish a method for the determination of 4 kind alkaloids in Wuji Wan. Methods: HPLC method was established. The chromatographic column was Diamonsil-C18. The mobile phase was a mixture of acetonitrile - 0. 05 tool. L^- 1 potassium dihydrogen phosphate (48:52, V/Vcontaining 0. 4 g sodium laurylsulfate per 100 mL). The flow rate was 1.0 mL-min-1, and the detection wavelength was 284 nm. Results: The calibration curves for berberine hydrochloride, epiberberine hydrochloric, coptisine hydrochloride, palmatine hydrochloride were linear in the concentration range of 5.0-50. 0, 0. 4- 4.0, 2.0-20.0, 2.0-20. 0 g. mL^-1, respectively. The average recoveries for berberine hydrochloride, epiberberine hydrochloric, coptisine hydrochloride, palmatine hydrochloride were 102. 4%, 100. 1%, 100. 0% , 102. 0% , respectively. Precision of the method was 0.36% , 1.50% , 1.60% , 0.86% (RSD, n = 6), respectively. Conclusion: The method can be used for quantitative determining of Wuji Wan.
