摘 要:目的 探讨老年小鼠淋巴结 T淋巴细胞的功能。方法 采用 3H- Td R掺入法测定淋巴细胞的增殖 ;3H-缬氨酸掺入法测定总蛋白合成 ;以依赖株细胞增殖反应测定法检测 IL - 2活性 ;3 H-缬氨酸掺入 IL- 2单克隆抗体 - IL - 2复合物 ,检测 IL- 2蛋白合成能力。结果 2 0月龄老年小鼠和 2 7月龄老年小鼠与 4月龄青年小鼠相比 ,其淋巴结细胞的增殖能力分别下降 65.6%和 32 .8% ;总蛋白合成下降 40 .2 %和 48.9% ;IL - 2活性下降 36.6%和 71 .5% ;IL- 2蛋白合成能力下降 32 .9%和 57.1 %。结论 老年小鼠淋巴结细胞对 Con A刺激的增殖能力、总蛋白合成水平、IL- 2活性及 IL-Objective It is well known that the immune functin declines with aging.Previous works in our laboratory showed that the production of IL 2,IL 3,GM CSF and NLK decreased in spleen lymphocytes from aged mice,and gene transcription of these factors was also inhibited.In this study,the functional changes in lymphocytes from lymph nodes of 27 months and 20 months aged mice were studied further.Methods The proliferation response of lymphocytes was determined by 3H thymidine incorporation. 3 H valine incorporation was used to determine protein synthesis capability.IL 2 activity was measured by CTLL 20 cell(IL 2 dependent cell line)proliferation.Results After ConA stimulation,the proliferation response of lymphocytes from lymph nodes of aged mice(27 month old and 20 month old)declined by 65 6% and 36 8%,resectively,as compared with young mice(4 month old).Total protein synthesis decreased by 40 2% and 48 9%.IL 2 activity declined by 36 6% and 71 5%.Meanwhile,IL 2 protein synthesis markedly decreased.Conclusions The results showed that the lymphocyte proliferation,total protein synthesis IL 2 activity and IL 2 protein synthesis from aged mice lymph nodes markedly decreased.These results demonstrate that lymphocytes from lymph nodes and spleen of aged mice has the similar function defects.
