摘 要:目的研究人类红细胞MN血型系统中抗-M的产生与相关基因GYPA表达的相关性,同时揭示M抗原的表达与抗-M产生之间的关系。方法选择无偿献血者和临床中发现的血浆中含有IgG、IgM或两者共存的16例抗-M血液样本,以血清学及分子生物学方法对样本进行MN血型检测,应用自行设计的一对特异性引物检测MN血型相关基因GYPA第二外显子的碱基序列。结果 16例样本中12例表现型为NN型,2例为MN型,2例为MM型。基因测序结果与GenBank参比序列比对,未发现突变碱基,血型表现型的基因定型与血清学定型相一致,经过谱细胞鉴定,该16例患者的血浆中全部携带抗-M抗体。结论人类红细胞MNS血型中抗-M的产生与M抗原是否存在没有必然关联,与相关基因GYPA的表达也无明显关联。Objective To study human MNS blood type system in the anti-M production and expression of related gene GYPA,and to reveal the relationship between expression of M antigen and anti-M antibody production.Methods To select donors and 16 blood samples whose blood plasma was found to contain IgG or IgM or coexistence of both,The MN blood types were checked by serological and molecular methods,A pair of primers were designed to detect the MN blood type gene GYPA of the second exon sequences.Results Twelve samples were tested for NN type,two samples for MN type by the gene sequencing.The genotyping method showed consistency of the results with the serologic test and no mutation was noted,The anti-M antibodies were found to exist in 16 blood samples by cells spectrum identification.Conclusion The anit-M production is uncorrelated to the presence of M antigen and the expression of GYPA gene.
