八宝丹对结肠癌荷瘤小鼠的血、脾和骨髓中的髓系抑制性细胞的影响  被引量:20

摘  要:目的:探讨中药八宝丹的抑瘤作用和肿瘤免疫学机制。方法:建立C26荷瘤小鼠模型,分别给予吉西他滨和八宝丹进行治疗。治疗后,计算各组抑瘤率,用流式细胞仪分别检测小鼠外周血、脾脏细胞悬液、骨髓细胞悬液中髓系抑制性细胞(MDSC)比例。结果:C26荷瘤小鼠血液、脾脏及骨髓中MDSC比例明显增高,与空白对照组小鼠比较有显著性差异(P<0.01)。吉西他滨和八宝丹都可以抑制肿瘤的生长,抑瘤率分别为51.21%和30.43%。与模型组相比,吉西他滨组能降低荷瘤小鼠外周血和脾脏中MDSC含量(P<0.01),而八宝丹能降低荷瘤小鼠外周血、脾脏和骨髓细胞中MDSC比例(P<0.05,P<0.01)。结论:八宝丹对荷瘤小鼠外周血、脾脏及骨髓中的MDSC比例有明显的降低作用,这可能是其抗肿瘤,改善肿瘤免疫的机制之一。Objective: To observe therapeutic effect and mechanism of Chinese medicine Babaodan to colon carcinoma. Methods: Mice models of C26 colon carcinoma were established, and treated with Gemcitabine or Babaodan. The inhibition rate of tumor was measured. The MDSC proportion in the blood, spleen and bone marrow were measured by flow cytometry. Results: The MDSC proportion of the blood, spleen and bone marrow in the C26 colon carcinoma mice significantly increased than that in the control group significantly (P〈0.01). Gemcitabine and Babaodan could inhibit tumor growth, and the inhibition rates were 51.21% and 30.43%, respectively. Compared with model group, Gemcitabine group could decreased the content of MDSC in the blood, spleen (P〈0.01), while Babaodan could reduce the MDSC proportion of the blood, spleen and bone marrow cell suspension (P〈0.05, P〈0.01). Conclusion: Babaodan can reduce the MDSC proportion of the peripheral blood, spleen, and bone marrow in C26 colon carcinoma mice, and improve the immunity inhibited by tumor, which might be the anti-tumor mechanisms of Babaodan.
