摘 要:目的:基于均匀设计法剖析益气化痰方防治抑郁症模型大鼠不同作用途径的效应中药配伍,探索中药复方多途径综合药理作用机制的研究方法和意义。方法:以益气化痰方中4味中药(半夏、陈皮、人参、茯苓)为研究对象,采用孤养加慢性轻度应激相结合诱导大鼠抑郁症模型,按均匀设计法"4因子6水平表"分组灌胃给药。观测大鼠下丘脑去甲肾上腺素(NE)、5-羟色胺(5-HT)、多巴胺(DA)水平,血清铜、锌以及血浆促肾上腺皮质激素(ACTH)、皮质醇(COR)含量,按均匀设计进行回归分析。结果:与对照组比较,模型组大鼠下丘脑NE、DA、5-HT水平明显降低(P<0.01,P<0.05),血浆ACTH、COR浓度明显升高(P<0.05,P<0.01),血清锌含量降低、铜含量升高(P<0.01)。应用均匀设计法结合药效学实验分析得出效应中药均是陈皮、人参、茯苓,而三者剂量配比有所不同,且与半夏相关性较小。结论:①益气化痰方中陈皮、人参、茯苓是作用于各途径的主要效应中药配伍,具有调节脑神经递质、微量元素水平、抑制内分泌亢进等多途径药理作用,半夏在这3条途径中均未发挥明显效应;②均匀设计法对于明确中药复方综合药理作用的物质基础(效应中药配伍)具有重要方法学意义。Objective: To analyze the most effective drugs of the compound traditional Chinese medicine prescription Yiqi Huatan Decoction (YQHTD) on rats model of depression on different pathways based on uniform design experimentation, and to explore the traditional Chinese medicine research method and significance of the multi-channel integrated pharmacological mechanism. Methods: Selected the four factor (tangerine peel, ginseng, Poria and Pinellia) in Yiqi Huatan Decoction as subject investigated. Rats model were made up by CUMS and life of separation, and grouped by table U6 × (64) in uniform design and gavaged with drugs. Observed the levels of hypothalamic monoamine NE, 5-HT and DA, and the content of serum copper, zinc, plasma ACTH and COR. Made regression analysis by uniform design. Results: Compared with control group animals, the levels of NE, DA and 5-HT in hypothalamus decreased significantly in depression rats (P〈0.05, P〈0.01), and the contents of ACTH and COR increased (P〈0.05, P〈0.01), and the serum copper level increased and the serum zinc level decreased significantly (P〈0.01). Applied uniform design combined with pharmacodynamic experiments to analyze the effects of traditional Chinese medicine were tangerine peel, ginseng, Poria, but in different ratio and having less relevant with Pinellia. Conclusion: ①YQHTD had multi-channel pharmacological effects in regulating brain neurotransmitters and trace element levels,suppressing endocrine overactivity. The effects of medicine acting on each pathway are mainly tangerine peel, ginseng, poria; and pinellia plays no obvious effect in these three pathways. ②Uniform design had important methodological significance in clarifying the material basis of general pharmacologic action of the compound traditional Chinese medicine (compatibility of the effects of traditional Chinese medicine).
