摘 要:目的 :观察养阴润肠方对慢传输型便秘大鼠结肠Cajal间质细胞的影响,探讨其通便的机制。方法 :随机将30只SD大鼠分为造模组(24只)及正常对照组(6只)。正常对照组给予蒸馏水灌胃,造模组采用大黄递增灌胃法制作慢传输型便秘大鼠模型,造模成功后,大鼠随机分为模型组、莫沙必利组、养阴润肠方组、自然恢复组。模型组及正常对照组予以一期处死,并采用活性炭灌胃法检测结肠传输功能、观察结肠内存留粪便粒数、实时荧光定量PCR法检测c-kit基因表达的变化。其他组继续给予相应干预措施干预30 d,之后检测上述指标。结果 :模型组结肠内存留粪便粒数较正常组多(P<0.01),养阴润肠方组及莫沙必利组结肠内存留粪便粒数较模型组少(P<0.01);模型组较正常组碳末推进率明显降低(P<0.01),与模型组比较,莫沙必利组及养阴润肠方组碳末推进率明显升高(P<0.01),而自然恢复组与模型组比较,没有统计学意义(P>0.05);模型组c-kit表达水平较正常组明显降低(P<0.01),恢复组与模型组比较,有恢复趋势,但无统计学意义(P>0.05),养阴润肠方组及莫沙必利组均有所改善(P<0.01,P<0.05),但莫沙必利组较养阴润肠方组表达水平低(P<0.05),莫沙必利组较恢复组表达量有所增高,但无统计学差异(P>0.05)。结论 :养阴润肠方具有较好的增强慢传输型便秘大鼠的结肠传输功能的作用,其通便机制可能为通过促进Cajal间质细胞的再生及修复而达到促进肠动力,从而实现通便的作用。Objective: To observe the effect of Yangyin Runchang Prescription on interstitial cells of Cajal of rats with slow transit constipation. Method: A total of 30 SD rats were used in the study, of which 6 were included in normal control group, and 24 were used to induce STC by intragastric administration of gradually increasing doses of rhubarb suspension. STC was successfully induced. The rats were randomized into model controls, Mosapride group, Yangyin Runchang Prescription group and recovery group. Model controls and normal control group were killed at the same time before medication administration. And then, the carbon powder propulsion rates in the intestine were measured and expressions of c-kit mRNA in ICC in the two groups were measured by Realtime PCR. After that, each medication 'administration team rats were given the corresponding drugs while recovery group was gavaged with the same volume saline for one month. The carbon powder propulsion rates in the intestine and the expression of c-kit mRNA in ICC were measured of these groups. Result: Carbon powder propulsion length in the model group was shorter than that of the normal group (P〈0.01 ) . Compared to the model control group, the carbon powder propulsion rates in Mosapride group and Yangyin Runchang Prescription group were faster significantly (P〈0.01), but there was no significant difference between the model group and recovery group. The expression of c-kit mRNA in ICC in the model group was lower than that of the normal group (P〈0.01 ), and there was no significant difference between the model group and recovery group (P〉0.05 ). The expression of c-kit mRNA in ICC in the Mosapride group was lower than that of Yangyin Runchang Prescription (P〈0.05 ). Conclusion:Yangyin Runchang Prescription can promote gastrointestinal motility and repair and regenerate the Cajal cells in rats to move the bowels.
