摘  要:目的 :探讨蜜环菌多糖含药血清对Aβ25-35诱导大鼠肾上腺嗜铬细胞瘤细胞(PC12细胞)损伤保护的作用机制。方法 :在Aβ25-35诱导PC12细胞损伤模型的基础上加入蜜环菌多糖含药血清,采用比色法检测含药血清对Aβ25-35诱导PC12细胞损伤培养液中乳酸脱氢酶的活力、丙二醛含量及超氧化物歧化酶活力的影响。结果 :不同浓度的含药血清对Aβ25-35损伤的PC12细胞的存活率有提高作用,含药血清浓度在5%时达最大值(P<0.05或P<0.01);5%浓度的含药血清可明显降低模型细胞培养液中LDH活力、MDA含量,提高SOD活力。结论 :蜜环菌多糖含药血清对Aβ25-35损伤的PC12细胞有保护作用,机制可能与提高细胞抗氧化能力有关。Objective: To study the protective mechanism of armillaria polysaccharide serum on injuries of PC12 cells induced by Aβ25-35. Methods: Based on the model of injuries of PCI2 cells induced by Aβ25-35' the models were treated with armillaria polysaccharide serum at different concentrations. MTT method was used to detect the influence of armillaria polysaccharide serum to PC12 cell proliferation inhibited by Aβ25-35. Colorimetric method was used to detect the influence of armillaria polysaccharide serum on activity of lactic dehydrogenase, MDA content and SOD activity in damaged PC 12 cell culture. Results: Different concentrations of armillaria polysaccharide serum increased the survival rate of damaged PC 12 cells induced by Aβ25-35, the peak was at 5% (P〈0.05 or P〈0.01 ); 5% concentration of armillaria polysaccharide serum can obviously reduce LDH activity, MDA content, and improve SOD activity in vitro. Conclusions: Armillaria polysaccharide can prevent PC 12 cell from Aβ25-35 injury, and the mechanisms of which may be related with the improved anti-oxidation ability of the cell.
