摘　　要：目的研究生长转化因子β(transforming growth factor beta,TGF-β)在血管紧张素Ⅱ(AngiotensinⅡ,AngⅡ)对体外培养的人成纤维细胞(HT-1080)toll样受体2(toll like receptor 2,TLR2)调控中的作用。方法取处于对数生长期的成纤维细胞,分为5组:空白对照组、阴性对照组,AngⅡ刺激组、TGF-β刺激组、TGF-β抑制剂Decorin+AngⅡ共刺激组。给药后采用实时定量PCR检测各组细胞内TLR2基因的表达量,并对各组结果进行统计分析。结果AngⅡ作用组中TLR2的表达量高于空白对照组(P<0.05);TGF-β组中低浓度(0.1、0.01μg/L)促进细胞内TLR2的表达,且表达量高于空白对照组,高浓度(1.0、10μg/L)抑制TLR2的表达,且表达量低于空白对照组(P<0.05);TGF-β抑制剂Decorin和AngⅡ联合作用组中TLR2的表达量高于空白对照组,且最高表达量高于其他各组(P<0.05)。结论 AngⅡ能够上调HT-1080中TLR2的表达量;TGF-β高浓度时抑制HT-1080中TLR2表达量,低浓度时促进TLR2的表达;在TGF-β低浓度时可与AngⅡ协同促进HT-1080中TLR2的表达。Objective To investigate the effect of transforming growth factor beta （ TGF-β） on the expression of toll like receptor 2 （ TLR2 ） regulated by angiotensinⅡ（ AngⅡ） in human fibroblast （ HT-1080 ） .Metho ds The cultured fibroblasts at logarithmic growth phase were divided into 5 groups：blank control group,negative control group,AngⅡgroup, TGF-βgroup,TGF-βinhibitor （ decorin ）＋AngⅡgroup.The expression levels of TLR2 mRNA were detected by RT-PCR in all the groups.The results were statistically analyzed .Results After treating HT-1080 with AngⅡ,the expression levels of TLR2 were significantly higher than those in blank control group （ P ＆lt;0.05）.The low-dose TGF-β（0.1,0.01 μg/L） increased the expression levels of TLR2, which were significantly higher than those in blank control group , however, the high-dose TGF-β（1.0,10μg/L） decreased the expression levels of TLR 2,which were significantly lower than those in blank control group （ P ＆lt;0.05）.The expression levels in TGF-βinhibitor （decorin ）＋AngⅡgroup were significantly higher than those in blank control group .Conclusion AngⅡcan up-regulate the expression levels of TLR 2.The high-dose TGF-βcan inhibit the expression of TLR2,however,low -dose TGF-βcan promote the expression of TLR2.The low -dose TGF-βin coordination with AngⅡcan increase the expression levels of TLR 2 in HT-1080 .
