益气消癥法中药含药血清干预ER mRNA及VEGFR mRNA表达抑制EA.hy926增殖的研究

摘　　要：[目的]探讨益气消癥法中药含药血清对人脐血管内皮细胞株(EA.hy926)增殖的影响,了解其抑制子宫肌瘤血管生成的途径。[方法]体外培养EA.hy926细胞,分为正常对照组、雌二醇(E2)组、E2加中药高、中、低剂量组,采用噻唑蓝(MTT)法、划痕法、流式细胞法、逆转录-聚合酶链反应(RT-PCR)技术检测细胞的增殖、迁移、凋亡率及ERαmRNA、ERβmRNA与VEGFR1mRNA、VEGFR2mRNA表达水平。[结果]益气消癥法中药可明显阻断E2对EA.hy926细胞增殖和迁移的促进作用,降低ERαmRNA与VEGFR1mRNA、VEGFR2mRNA表达,阻断E2对EA.hy926细胞的凋亡的抑制作用,上调ERβmRNA的表达,与E2组比较差异有统计学意义(P<0.01),且作用效果呈剂量相关性。[结论]该法方药可通过下调ERαmRNA与VEGFR1mRNA、VEGFR2mRNA表达,上调ERβmRNA的表达,从而抑制EA.hy926细胞的增殖和迁移,促进其凋亡,干预血管生成是其治疗子宫肌瘤的作用机制之一。[Objective] To observe the effect of Yiqi Xiaozhengcontained serum on proliferation on EA.hy926 cells from umbilical vein of human and understand the way of inhibiting angiogenesis in human uterine myoma. [Methods] EA.hy926 cells cultured in vitro were di vided into six groups： normal group, E2 group, E2 and ICI group, E2 and traditional Chinese medicine of high, middle, low dose group. EA. hy926 cells proliferation, migration ability, apoptosis rate, ERmRNA, ERI3mRNA and VEGFRlmRNA, VEGFR2mRNA expression were detected by MTT, scratch method and flow cytometry and RTPCR technology respectively. [Results] Yiqi Xiaozhengcontained serum could blockade the promoting effect of E2 on the proliferation and migration of EA.hy926 decrease the expression of ERβmRNA, VEGFRlmRNA and VEGFR2mRNA, blockade the inhibitory effect of E2 on the apoptosis of EA.hy926 cells, upregulate ERI3mRNA ex pression with a significant difference when compared with E2 group （P〈0.01）. The effect and dose were dosedependent. [Conclusion] Yiqi Xiaozhengcontained serum can inhibit proliferation and migration and promote apoptosis of EA.hy926 cells through downregulating the expression of ERccmRNA, VEGFRlmRNA and VEGFR2mRNA and upregulating the expression of ERβmRNA. Intervention of an giogenesis is one of the mechanisms for the treatment of uterine myoma.
