摘  要:目的:通过构建HCN4腺病毒载体,以大鼠骨髓间充质干细胞(MSCs)为基因转染的平台,观察HCN4外源基因对MSCs转染效果及其功能表达。方法:携带HCN4腺病毒载体体外转染MSCs,测定转染效率,免疫荧光法测定HCN4蛋白表达,膜片钳全细胞记录If电流。结果:携带HCN4腺病毒载体体外转染MSCs后可检测到HCN4蛋白及If电流表达。结论:携带HCN4腺病毒载体转染大鼠MSCs后可使MSCs表达HCN4蛋白及If电流,可为生物心脏起搏治疗提供合适的靶基因及种子细胞。Objective:To construct the rat MSCs transferred by Ad-HCN4 is tested whether protein of HCN4 in MSCs are expressed and If current is generated. Methods:The fluorescence microscope was used to test the transfection rate of the rat MSCs transferred by Ad-HCN4. The HCN4 protein was detected by the cellular immunofluorescence. The patch clamp whole cell record model was adopted to record the If current of the MSCs transferred with the target gene. Results: The rat MSCs transferred by Ad-HCN4 express HCN4 protein. The If current of the MSCs was confirmed by the patch clamp. Conclusions: The rat MSCs transferred by Ad-HCN4 can express HCN4 protein. The If current of the MSCs is confirmed by the patch clamp. On the whole, HCN4 and MSC are the adaptive target gene and seed cells for the biopacemaker.
