摘 要:目的:观察替米沙坦对代谢综合征患者内皮祖细胞(EPCs)功能的影响。方法:将代谢综合征患者76例随机分为常规组和试验组各38例,同时选择健康体检者38例为对照组。试验组在常规用药的基础上加服替米沙坦80 mg/d,在治疗前及治疗后4周取外周血,密度梯度离心法分离培养EPCs,免疫荧光双染法鉴定EPCs,流式细胞仪、30 min贴壁法观察EPCs的体外增殖、粘附能力,Griess法检测一氧化氮(NO)分泌量,RTPCR半定量测定内皮型一氧化合酶(eNOS)基因表达。结果:与常规组相比,治疗后试验组EPCs的体外增殖、粘附能力明显增强,NO分泌量明显增多,eNOS基因表达明显上调,P均<0.05;而常规组相比则无明显改变。结论:替米沙坦能改善代谢综合征患者EPCs功能,这与上调eNOS基因表达,促进NO分泌有关。Objective: To observe the effect of Telmisartan on the endothelial progenitor cells (EPCs)' function of patients with metabolic syndrome (MS). Methods: 76 patients with MS were randomly divided into conventional group (n=38) received conventional treatment and experiment group (n=38) received Telmisartan (80 mg/d) on the basis of conventional treatment. Before and 4 weeks after treatment, EPCs were isolated from blood of patients by density gradient centrifugation and were cultured.The proliferation, adhering ability and the NO level were assayed by flow cytometric analysis, adhesion test and Griess method after identification. RT-PCR was performed to determine the eNOS expression. Results: Compared with conventional group, the proliferation, adhesion ability and NO production were significantly increased, and eNOS expression was significantly raised after treatment in the experiment group, all P 〈 0.05. These changes were not found in conventional group. Conclusions: Telmisartan can improve the EPCs' function of patients with MS by increasing NO production based on raised eNOS expression.
