用于家蚕转基因的二化性品种蚕卵的预处理研究

摘  要:从二化性品种获得优质的非滞育蚕卵作为显微注射受体,一直是家蚕转基因技术中亟待解决的问题之一.目前家蚕转基因技术中非滞育蚕卵主要是通过完全低温暗催青、早浸酸2种方法得到,但这2种方法分别存在催青时间长、浸酸时间难以把握的局限.对完全低温暗催青技术进行了改进,即在家蚕胚胎发育至温度敏感的缩短期(戊3期)后再对蚕卵低温黑暗处理直至孵化,结果表明:后期低暗组能有效地获得非滞育蚕卵并且较完全低暗组催青时间缩短3~4天.并进一步分析了后期低暗组、完全低暗组以及早浸酸组3种处理得到的非滞育卵的健康性,结果表明后期低暗组和完全低暗组健康性差异不大,均明显优于早浸酸组;后期低温暗催青在蚕卵孵化所需时间上具有明显的优势,且获得的非滞育卵健康性亦可得到保证,有望成为人们获得非滞育卵的一条有效途径.Accessing to high-quality non-diapause silkworm eggs as the microinjection receptor is one of the problems to be solved in the silkworm transgenic technology.Currently,there are two methods that can provide non-diapause eggs.One is whole low-temperature-dark incubation and the other is early acid treatment.But they have their disadvantages:the former is time-consuming and the pickling time is difficult to control for the latter.A modified low-temperature-dark incubation technology is proposed in this paper, i.e.low-temperature-dark incubation is postponed until the temperature-sensitive period of embryo development-head thorax differentiation.The result shows that the new method can acquire non-diapuase silkworm eggs effectively and save 3-4days compared with the whole low-temperature dark incubation.Analysis of the health of the non-diapause silkworm eggs obtained with the three methods shows that the health of the silkworm eggs from the new method is better than that from acid treatment.It is concluded that low-temperature dark incubation at a later development stage has obvious advantages.The new method is expected to provide an effective way for people to get non-diapuase eggs.
