超低温冷冻对德国牧羊犬顶体酶类活性的影响

摘  要:为了探讨超低温冷冻对德国牧羊犬透明质酸酶(HYD)、顶体酶(ACE)和酸性磷酸酶(ACP)活性的变化,试验分别对冷冻前后德国牧羊犬精子采用改良明胶底膜法、改良BNPNA法、磷酸苯二钠法对精子HYD、ACE和ACP活性进行研究。结果表明:超低温冷冻前后精子HYD、ACE和ACP活性极显著降低(P<0.01);解冻后精子3种顶体酶类活性分别与精子活率和顶体完整率呈正相关,其中ACE活性与顶体完整率呈极显著正相关,相关系数为0.487(P<0.01)。3种顶体酶类活性与畸形率均呈显著负相关(P<0.05),ACE活性与精子顶体完整率的相关系数为0.487,相关度大于HYD(0.439和0.459)和ACP(0.440);而HYD活性与精子活率的相关系数为(0.404和0.423),相关度大于ACE(0.377)和ACP(0.369)。说明超低温冷冻使德国牧羊犬精子3种顶体酶类活性均显著降低;3种顶体酶类活性分别与精子活率和顶体完整率呈正相关,与畸形率呈负相关。To investigate the effects of cryopreservation on the activities of hyaluronidase (HYD), acrosome enzyme (ACE), and acid phosphatase (ACP) in German shepherd dog, the modified gelatin - substrate film technique, the modified BAPNA method , and disodium phenyl phosphate method were used to determine the sperm HYD, ACE and ACP activities of German shepherd dog before and after freezing, respectively. The results showed the sperm HYD, ACE and ACP activities were significant reduced (P 〈 0.01 ) both before and after cryopreservation. The activities of the three kinds of acrosomal enzymes for the thawed sperm were positively correlated with sperm motility and acrosome integrity, in which the ACE activity had a highly significant positive correlation ( P 〈0.01 ) with the acrosome integrity and the correlation coefficient was r = 0. 487. The activities of the three kinds of acrosomal enzymes had highly significant negative correlations ( P 〈 0.05 ) with the aben'ation rate. The correlation coefficient between the ACE activity and sperm acrosome integrity was 0. 487, which was greater than that of HYD (0. 439 and 0. 459 ) and ACP(0.440), while the correlation coefficients between the HYD activity and the sperm motility rate were 0. 404 and 0. 423 respectively, which were greater than those of ACE (0. 377 ) and ACP (0. 369) respectively. The results indicate that the HYD, ACE and ACP activities are significantly reduced by cryopreservation. The activities of the three kinds of acrosomal enzymes have positive correlations with the sperm motility and acrosome integrity, showing negative correlations with the aberration rate.
