摘 要:目的优化延胡索有效成分的提取、纯化工艺。方法以提取物中延胡索乙素量为考察指标,在单因素实验基础上,采用正交试验的方法对影响延胡索有效成分提取、纯化的因素进行优化。结果最佳提取工艺为:药材粗粉(10目)于60℃加12倍量水浸泡3次,每次30 min;最佳纯化工艺为:提取液用药材1.5倍量的WLD大孔吸附树脂吸附,3倍树脂量的70%乙醇洗脱。结论优化的提取、纯化工艺简单、稳定且能有效保证延胡索乙素不被破坏。Objective To optimize the xtraction and purification technological condition of active ingredients in Corydalis. Meth- ods Using the amount of tetrahydropalmatine as indicator, on the basis of single - factor experiments, the extraction and purifica- tion technological conditions of alkaloids from Corydalis were optimized by orthogonal design - method. Results The extraction technical of Corydalis is: the coarse powder( 10 mesh) soaked by 12 times amount of water for 30 rain and 3 times in 60~C ;The purification technical is : water - decocted solution from Corydalis were adsorbed by 1.5 times WLD - resin of drug , the effective fractions of Corydalis were desorbed by 3 times 70% ethanol of wet resin volume. Conclusion The optimized extraction technology is stable and feasible and guaranteeing tetrahydropalmatine not to be destroyed
