针刺对哮喘大鼠BALF中嗜酸性粒细胞及肺组织p38MAPK的影响

摘　　要：目的观察针刺对支气管哮喘大鼠支气管肺泡灌洗液中嗜酸性粒细胞,肺组织p38蛋白激酶(p38MAPK)蛋白表达的影响,探讨针刺对哮喘大鼠气道炎症的改善机制。方法将SPF级雄性SD大鼠随机分为空白组(C组)、哮喘模型组(M组)、哮喘模型针刺组(A组),每组10只。制作卵蛋白致敏哮喘大鼠模型,取大椎、肺俞(双)、风门(双)穴位给予针刺,隔日一次,共针7次。针刺干预实验结束时,计数哮喘大鼠支气管肺泡灌洗液(BALF)中嗜酸性粒细胞;用Western Blot技术检测大鼠肺组织p38MAPK的蛋白含量。结果 M组大鼠BALF中嗜酸性粒细胞数较C组显著增多(P<0.05);A组大鼠BALF中白细胞和嗜酸性粒细胞数明显低于M组(P<0.05)。M组大鼠肺组织p38MAPK蛋白表达较C组增多(P<0.05);A组大鼠肺组织p38MAPK蛋白表达低于M组(P<0.05)。结论针刺对哮喘的治疗作用与降低大鼠BALF中嗜酸性粒细胞数目,降低肺组织p38MAPK蛋白表达有关。Objective To investigate the effects of acupuncture on eosinophils in bronchoalveolar ]avage fluid （BALF） and pro- tein expression of p38MAPK in lung tissue of asthmatic rats to explore the anti - inflammatory mechanisms of acupuncture on asth- matic rats. Methods The SPF male SD rats were randomly divided into normal control group （ C group）, asthma model group （ M group）, and the acupuncture treatment group （ A group）, with 10 rats in each group. Asthma rat models were established by sen- sitization with ovalbumin. GVI4 （Dazhui）, Bilateral BL13 （Feishu）, and Bilateral BL12 （Fengmen） of the rats in A group were located and aeupunetured once every other day for two weeks. At the end of the experiment, eosinophils were counted in BALF ofasthma rats; Western Blot was employed to observe the protein expression of p38MAPK in lung tissue of rats. Results Compared with that in C group, the count of eosinophil in BALF of rats in M group was significantly increased （ P 〈 0.05 ） ; while compared with that in M group, the count of eosinophil in BALF of rats in A group was significantly decreased （ P 〈 0.05 ）. Compared with that in C group, protein expression of p38MAPK was increased in M group （P 〈 0.05 ） ; while compared with that in M group, protein expression of p38MAPK was increased in A group （P 〈 0.05 ）. Conclusion Acupuncture can reduce the count of eosino- phils in the BALF and reduce the protein expression of p38MAPK in lung tissue, which may he partly related to the role of acu- puncture in the treatment of asthma.
