摘  要:目的探讨1,25-(OH)2-D3对进展性肾炎大鼠肾小管间质胶原蛋白Ⅲ(ColⅢ)表达的影响。方法将实验动物按随机数字表法分为3组(n=7):假手术组(对照组)、进展性肾炎组(模型组)、6 ng/(100 g·d)1,25-(OH)2-D3干预组(治疗组)。除对照组外,其余组均采用右肾切除并2次注射IgG(OX-7)鼠抗人Thy1.1单克隆抗体,制备进展性肾炎大鼠模型。1,25-(OH)2-D3干预8周后,检测各组大鼠24 h尿蛋白、血肌酐(SCr)、尿素氮(BUN)及电解质的变化,Masson染色观察肾组织病理改变。免疫组织化学检测肾小管间质ColⅢ的表达。结果与模型组比较,1,25-(OH)2-D3治疗不仅使大鼠24 h尿蛋白及BUN均明显降低(P<0.05),还能使ColⅢ表达显著下调(P<0.05)。肾组织病理损害明显减轻。结论 1,25-(OH)2-D3可能通过明显降低进展性肾炎大鼠肾小管间质ColⅢ的表达,减轻肾小管间质纤维化而发挥肾脏保护作用。Objective To investigate the effects of 1,25-(OH)2-D3 on the expression in tubulointerstitial of Col Ill in rat with progressive nephritis. Methods Experimental rats were divided into three groups according to random number table method:sham operation group (control group),progressive nephritis group (model group),and progressive nephritis group treated with 6 ng/(100 g-d) 1,25-(OH)2-D3 (treatment group).In addition to the control group,the other progressive nephritis model was established by right nephrectomy and injection with IgG(OX-7) mouse monoclonal anti-Thyl.1 an- tibody twice.Progressive nephritis rats received 1,25-(OH)2-D3 by intragastric administration for 8 weeks,the urine pro- tein of 24 hours,serum creatinine (SCr),blood urea nitrogen (BUN)and the levels of calcium and phosphorus in rats of each group were measured.The renal morphologic changes were evaluated on Masson stained sections.The expression in tubulointerstitial of Col 11I was detected by immunohistochemical. Results The 24 hours urine protein and BUN in tubulointerstitial were lower in 1,25-(OH)2-D3 treatment group than those in model group (P〈0.O5).In addition,the expressions of Collll mRNA in 1,25-(OH):-D3 treatment group were significantly lower than those in model group (P〈0.05). The renal pathological damage was significantly reduce. Conclusion 1,25-(OH)2-D3 may protect kidney by de- creasing the expression of tubulointerstitial fibrosis gene Col III in the progressive nephritis model rats,and reduce renal tubular interstitial fibrosis and renal protective effect into full play.
