Effect of Qufeng Tongluo Recipe on Expression of Desmin and CD2AP Proteins in Adriamycin-induced Nephropathy Rats:an Experimental Research

摘  要:目的探讨祛风通络方对阿霉素肾病大鼠足细胞结蛋白(Desmin)和CD2相关蛋白(CD2-associated protein,CD2AP)的作用。方法一次性尾静脉注射阿霉素制备肾病模型。3周后造模成功,随机分为空白对照组(A组,12只),模型对照组(B组,8只),祛风通络方小、中、大剂量组(C组、D、E每组8只)和阳性对照组(F组,8中)。从第4周起,A组和B组给生理盐水,C^E组分别给祛风通络方1.0、2.1、4.2 g/mL,F组给泼尼松25 mg/kg。各组均灌胃给药,10 mL/kg,1次/天,连续28天。期间进行24 h尿蛋白定量、血清生化指标检测,实验结束时电镜观察超微结构、Real-time PCR法及Western blot法检测足细胞骨架蛋白Desmin mRNA、裂孔隔膜蛋白CD2AP mRNA及其蛋白表达。结果 (1)与B组比较,C、D、E、F组24 h尿蛋白定量明显降低,差异有统计学意义(P<0.05)。(2)与B组比较,C、D、E组ALB均有所升高,差异有统计学意义(P<0.05);TC含量明显降低,差异均有统计学意义(P<0.05);F组TG反而有明显增加,差异有统计学意义(P<0.05)。(3)电镜观察发现,D、E、F组与B组比较,足细胞形态均有不同程度改善,特别是足突结构、足突数目改善明显,其D、E组改善更明显。(4)阿霉素肾病大鼠中Desmin mRNA、CD2AP mRNA及蛋白表达均升高,差异有统计学意义(P<0.05)。干预后,与B组比较,C、D、E、F组Desmin mRNA和CD2AP mRNA表达明显降低,差异有统计学意义(P<0.05)。(5)与A组比较,B组Desmin、CD2AP蛋白表达显著升高,差异有统计学意义(P<0.05)。与B组比较,C、D、E、F组CD2AP蛋白表达明显降低,D、E、F组Desmin蛋白表达明显降低,差异均有统计学意义(P<0.05);各剂量组间比较,C、D、E组Desmin及CD2AP蛋白表达逐渐降低,差异有统计学意义(P<0.05);与F组比较,C、D组CD2AP蛋白表达明显高,E组CD2AP蛋白表达明显低,差异有统计学意义(P<0.05);C、D、E组Desmin蛋白表达较高,差异有统计学意义(P<0.05)。结论祛风通络方对阿霉素肾病大�Objective To investigate the effect of Qufeng Tongluo Recipe (QTR) on the expres- sion of desmin and CD2-associated protein (CD2AP) in adriamycin-induced nephropathy rats. Methods The adriamycin-induced nephropathy rat model was induced by a disposable intravenous injection of adri- amycin. The model was successfully established after 3 weeks. Rats were then randomly divided into the blank control group (Group A, n =12), the model control group (Group B, n =8), the small, medium, large dose QTR group (Group C, n =8; Group D, n =8; Group E, n =8), and the positive control group (Group F, n =8). From the fourth week normal saline was given to rats in Group A and Group B, QTR 1.0g/mL, 2.1 g/mL, and 4.2 g/mL was respectively administered to those in Group C, D, and E. Prednisone 25 mg/kg was given to rats in Group F. All medication was performed by gastrogavage at 10 mL/kg, once daily, for 28 successive days. 24-h urinary protein excretion and sera biochemical indices were deter- mined during medication. At the end of the experiment, ultrastructure was observed, mRNA expression of desmin, mRNA and protein of CD2AP were detected by Real-time PCR and Western blot. Results (1) Compared with Group B, 24-h urinary protein excretion significantly decreased in Group C, D, E, and F (P 〈0.05). (2) Compared with Group B, AIb in Group C, D, and E increased (P 〈0, 05) and TC significantly decreased (P 〈0.05). TG significantly increased in Group F (P〈0.05). (3) Results of elec- tron microscope showed, compared with Group B, the morphology of foot cells was improved to various degrees in Groups D, E, and F, especially the foot process structure and the number of foot processes were significantly improved, which was more obviously shown in Group D and Group E. (4) mRNA ex- pression of desmin, mRNA and protein of CD2AP increased in adriamycin-induced nephropathy rats (P 〈 0.05).
