摘  要:目的:探讨急性心肌梗死(AMI)大鼠中缓激肽B1(BDKB1R)的表达及依那普利氯沙坦药物治疗对其的影响。方法:结扎大鼠左冠状动脉前降支制备心肌梗死模型。将60只大鼠随机分为假手术组10只,AMI组30只,干预组20只。AMI组分别于6h、24h、3w时取心肌梗死组织,分为6hAMI组、24hAMI组、3w AMI组,每组10只。将干预组分为3w依那普利干预组(A组)10只、3w氯沙坦干预组(B组)10只。分别采用免疫组织化学法检测AMI区BDKB1R蛋白,RT-PCR法检测BDKB1R mRNA的表达。结果:各组大鼠梗死区心肌BDKB1RmRNA和蛋白质的表达均差异有统计学意义,梗死后6h梗死区心肌BDKB1R mRNA和蛋白的表达增加,在24h时达到高峰,3w时下降。3wAMI组和A组、B组BDKB1RmRNA和蛋白的表达仍高于假手术组,但A组和B组高于3wAMI组。结论:心肌梗死后BDKB1R的表达增加,依那普利和氯沙坦可上调其表达,可能是其在心肌梗死中起到保护心肌作用的机制之一。Objective:To investigate the expression of BDKB1Rin acute myocardial infarction(AMI)rats and to study the effect of enalapril and losartan on it.Method:The rats model of AMI was induced by ligation of the left coronary artery.Rats were randomly divided into sham operation group,AMI group and pravastatin group.AMI group was divided into 6hMI group,24hMI group and 3wgroups,each group has 10rats.Pravastatin group was divided into three weeks enalapril group(A group),three weeks losartan group(B group).The expressions of BDKB1Rprotein and mRNA were detected by immunohistochemisty and RT-PCR respectively.Result:The expressions of BDKB1RmRNA and protein in the above groups were statistically different.The above indexes all exhibited an ascending tending at 6hafter AMI,and reached peak value at 24h,then descended at 3week.The above indexes in 3wgroup,A group and B group were still higher than those in sham operation group,and A group and B group were higher than 3wgroup.Conclusion:The expression of BDKB1Rincreases after myocardial infarction.Enalapril and losartan may regulate the express of BDKB1R,which may be one of the mechanisms to protect myocardium after AMI.
