摘 要:目的研究耐亚胺培南铜绿假单胞菌外膜蛋白(OprD2)缺失情况。方法取临床分离的铜绿假单胞菌49株,根据其耐药性分为亚胺培南耐药组(35株)和敏感组(14株),采用PCR方法检测各菌株的OprD2基因。采用MLVA方法对菌株进行基因分型,分析菌株之间的亲缘关系。结果 49株菌株中,共有8株OprD2基因PCR扩增阴性,缺失率为16.3%(8/49),其中耐药株缺失率为17.1%(29/35),敏感株缺失率为14.3%(2/14),差异无统计学意义(P>0.05)。MLVA基因分型无成簇菌株存在。结论广东中山地区临床分离株耐亚胺培南铜绿假单胞菌OprD2缺失率低,对亚胺培南耐药的作用不显著。MLVA分型表明当地尚未发生耐亚胺培南铜绿假单胞菌流行。Objective Study of the deletion situation of OprD2 gene of Pseudomonasaeruginosa (PA). Methods This study included 49 PA samples isolated from patients in hospital. The relationship between the mutation of OprD2 and drug resistance of the bacteria was investigated in 35 strains of imipenem (IMP) and 14 strains of IMP-sensitive PA. Multiple locus variable-number tandem repeat analysis (MLVA) method was used to analyze the relationship of the strains. Re- suits 8 of 49 strains (8/49, 16.3%) were shown to be negative for OprDz gene by PCR method. Among them, 17.1% (29/35)of the IMP-resistance strains were positive, and while 14.3 % (2/14) of the IMP-sensitive strains were positive. The result of Chi-square test revealed that the difference between the positive rates of the OprD2 gene in the IMP-resist- ance and IMP-sensitive PA was not statistically difference. MLVA result showed there were not clustered strains. Con- clusion The deletion rate of OprD2 gene was low in Zhongshan, Guangdong Province. MLVA result indicated that there was not the epidemic outbreak of the PA.
