摘 要:目的探讨新风胶囊治疗类风湿关节炎抗血管新生的可能作用机制。方法 50只SD大鼠随机为正常对照组、模型对照组、甲氨蝶呤组、雷公藤多苷片组、新风胶囊组,每组10只。除正常对照组外,其余各组均采用弗氏完全佐剂建立佐剂关节炎大鼠模型。造模后第19天,新风胶囊组给予新风胶囊溶液2.4 g/kg灌胃,雷公藤多苷片组给予雷公藤多苷片溶液10 mg/kg灌胃,均每天1次;甲氨蝶呤组给予甲氨蝶呤溶液3mg/kg灌胃,每周1次;正常对照组、模型对照组给予1.0g/kg生理盐水灌胃,每天1次,各组均灌胃30天。比较各组大鼠足跖肿胀度、关节炎指数及滑膜组织缺氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)的表达,并进行滑膜微血管密度(MVD)计数。结果与正常对照组比较,模型对照组大鼠足跖肿胀度、关节炎指数升高,滑膜组织MVD计数、VEGF、HIF-1α表达显著升高(P<0.01);与模型对照组比较,甲氨蝶呤组、雷公藤多苷片组及新风胶囊组大鼠足跖肿胀度、关节炎指数降低,滑膜组织MVD计数、VEGF、HIF-1α表达显著降低(P<0.05或P<0.01);并且新风胶囊组HIF-1α较甲氨蝶呤组、雷公藤多苷片组降低明显,VEGF较甲氨蝶呤组降低明显(P<0.05)。结论新风胶囊通过降低大鼠滑膜组织VEGF、HIF-1α的表达发挥抑制滑膜血管新生的作用。ABSTRACT Objective To research the possible antiangiogenic mechanism of Xinfeng Capsules (新风胶囊) for rheumatoid arthritis (RA). Methods Fifty SD rats were randomized into the normal control group, model control group, Methotrexate group, Tripterygium Glycosides Tablets group and Xinfeng Capsules (新风胶囊) group, with 10 in each. The adjuvant arthritis rat models were established with Freund's complete adjuvant. After modeling, the Xin- feng Capsules (新风胶囊) group was gavaged with 2.4mg/kg Xinfeng Capsules (新风胶囊) solution once claily. The Tripterygium Glycosides Tablets group was gavaged with 10 mg/kg Tripterygium Glycosides Tablets soluhon once daily. The Methotrexate group was gavaged with 3 mg/kg Methotrexate solution once weekly. The normal control group and model control group were gavaged with 1.0 mg/kg normal saline once daily. The intragastric administration lasted for 30 days. The plantar swelling degree and arthritic index was measured. The expressions of hypoxia-inducible fac- tor-1α (HIF-1α) and vascular endothelial growth factor (VEGF) were observed. The synovial microvessel density (MVD) was counted. Results Comparing with the normal control group, the plantar swelling degree, arthritic in- dex, MVD, VEGF expression and HIF-1α expression was significantly increased in the model control group (P 〈 0. 01 ). Comparing with the model control group, the plantar swelling degree, arthritic index, MVD, VEGF expres- sion and HIF-1α expression was significantly decreased in the Methotrexate group, Tripterygium Glycosides Tablets group and Xinfeng Capsules (新风胶囊) group (P 〈0. 05 or P 〈0. 01 ), especially HIF-1α expression in the X Capsules (新风胶囊) group as compared with the Methotrexate group and VEGF expression in the Xinfeng Capsules (新风胶囊) group as compared Conclusion Xinfeng Capsules (新风胶囊) has an antiangiogenic effect HIF-1α in synovial tissue.
