摘  要:WRKY是植物体内最大的转录因子家族之一,其成员在植物生长发育和抗逆过程中发挥重要作用。为了解水稻WRKY的功能,本研究利用蛋白质免疫印迹(w estern blotting,WB)技术,检测了8个WRKY转录因子在水稻叶片生长和Xa21介导的抗白叶枯病过程中的表达丰度变化。结果表明:OsWRKY12、43、55和86在苗期表达量较低,随叶片的生长表达逐步增加;OsWRKY50和84在苗期叶片中的表达量相对较高,成熟期下降;未检测到OsWRKY40/64和52在叶片中的表达。在Xa21介导的白叶枯病抗性反应中,OsWRKY40/64、50和52受白叶枯病菌诱导表达,OsWRKY84受侵染后表达量上调,OsWRKY12、43、55和86在抗病过程中没有检测到表达丰度的变化。进一步比较这些转录因子在抗、感和对照(Mock)反应中的表达,发现抗、感反应间蛋白质的表达变化是相似的。研究结果提示:OsWRKY12、43、55和86可能在叶片正常生长中发挥作用;OsWRKY40/64和52可能在水稻-白叶枯病菌互作过程中发挥作用;OsWRKY50和84则在叶片生长和抗白叶枯病过程中都有一定的功能。As one of the largest transcription factor families in plant, many WRKY mumbers were reported to play essential roles in the developmental process and responses to biotic/abiotic stress. To better understand the function of rice WRKY proteins, the expression profile of eight WRKY proteins in leaf growth and Xa21-media- ted Xanthomonas oryzae pv. oryzae (Xoo) resistance response was surveyed by western blotting (WB). The re- suits demonstrated that OsWRKY12, 43, 55 and 86 were expressed in a low level at seedling stage and increased gradually in later stages. OsWRKY50 and 84 were expressed in a high level at seedling stage, while decreased in the later stages. However, the expression level of OsWRKY40/64 and 52 was not detected in rice leaves at all stages. In the fice-Xoo incompatible interactions, the expression of OsWRKY40/d4, 50 and 52 was induced dra- matically, and that of OsWRKY84 was also increased. While the expression levels of OsWRKY12, 43, 55 and 86 were stable. Comparison analysis among rice-Xoo incompatible, compatible and mock treatments revealed a parallel expression pattern between the incompatible and compatible interactions. Based on these experiments, we postulated that OsWRKY12, 43, 55 and 86 might play a role in rice leaf growth, OsWRKY40/64 and 52 mightbe essential in the interaction between rice and Xoo, while OsWRKY50 and 84 might be involved in both proces- ses.
