摘 要:基于肝素钠对吖啶橙的共振散射增强作用,建立了一种测定肝素钠的新方法。在1.0mol/LHCl介质中,吖啶橙与肝素钠反应形成缔合物,使溶液共振散射(RS)增强,且在323nm波长处产生1个较强散射峰。在0.020~2.0μg/mL范围内,体系共振散射信号与肝素钠浓度之间有较好的线性关系,其检出限为0.185ng/mL。该方法灵敏、稳定、选择性好,可直接用于肝素钠注射液中肝素钠含量的测定,回收率为95.6%~102.99/6。A new analytical method has been developed based on the enhancement of resonance scattering of heparin sodium to acridine orange. In 1.0 mol/L HC1 solutions, acridine orange reacted with heparin sodium to form ion-association complexs, which made the resonance scattering intensity enhance obviously and created the maximum resonance scattering peak at 323 nm. Under the optimum experimental conditions, there was a good linear relationship between the resonance scattering intensity of the system and the heparin sodium concentration in the range of 0. 020-2.0μg/mL, and the detection limit was 0. 185 ng/L. This method has been applied to determine the heparin sodium content in heparin sodium injection with the recovery in the range of 95.6%-102.9%.
