摘  要:目的了解2012年甲型H1N1流感病毒HA基因和抗原性特征。方法对2012年分离的5株甲型H1N1流感病毒的血凝素(HA)进行测序,用软件进行基因种系进化树分析;对2株2012年的甲型H1N1和A/Sichuan/1/2009(H1N1)及2株2010年甲型H1N1流感病毒抗原性比较分析;1株2012年毒株与2009年我国代表毒株A/Sichuan/1/2009(H1N1)一起对2012年采集的169份血清进行HI试验,比较其HI抗体水平。结果 2012年甲型H1N1流感病毒的血凝素(HA)基因的碱基序列发生了1.11%的变异,氨基酸序列发生2.0%残基替换,主要是抗原性位点Sb和Ca1的变异为主。与我国代表株A/Sichuan/1/2009(H1N1)和成都的2011年的2株甲型H1N1HI实验比较,抗原性有差异。结论 2012年甲型H1N1的HA发生了一定程度的变异,主要在抗原性位点Sb和Ca1处发生了氨基酸替换,使得2012年甲型H1N1流感病毒的抗原性与2009年相比已经也发生了改变。Objective To study the hemagglutinin (HA) genetic and antigenic variation of influenza A (H1N1) viruses isolated in 2012. Methods Sequenced the hemagglutinin complete gene of pandemic influenza A (HIN1) viruses isolated in 2012. Compared antigen of two influenza A (H1N1) viruses isolated in 2012 with A/Sichuan/1/2009 (H1N1) and A (H1N1) viruses isolated in 2010. Tested the hemagglutination inhibition antibody of 169 serum sample collected in 2012 by A (H1N1) virus isolated in 2012 and A/ Sichuan/1/2009 (H1N 1). Results Compared with pandemic H1N1 virus HA sequence, there were 1.11% variation in gene and 2.0% change in amino acid residues. Main changes occurred at Sb and Cal antigenic sites. Compared with the China representative strain (A/Sichuan/1/2009 (H 1N 1) and the H 1N 1 strains isolated in 2011 by hemagglutinin inhibition test (HI), the antigenic differ- ences were significant. Conclusion Compared with (H1N1) HA in 2012, there are changes in the HA gene, and the main changes of amino acid in the different antigens in Sb and Cal, which results in changes of antigenicity in H1N1 in 2009 and 2012.
