摘　　要：目的探讨凝聚胺微板法在献血者不规则抗体筛查的规范试验。方法选用效价为16的IgG抗-D的人源血清,以传统凝聚胺试管法为基础,运用正交原理研究不同血清量、不同LIM液量对红细胞凝集的影响,并测定观察结果,获得最适宜的震荡参数,同时比较微板法、凝聚胺试管法灵敏度。结果选用1 000 r/min,震荡10 s,50μL血清、150μL LIM的反应体系,凝聚胺微板法抗体筛查特异性96.87%,灵敏度98.44%。结论凝聚胺微板法能够代替传统的凝聚胺试管法进行抗体筛查,且能批量操作,易于自动化。Objective The polybrene microplate,a standard method for blood donors＇irregular antibody screening was established. Methods The IgG anti-D anthropogenie serum was selected,which their titer was 16. Based on traditional po- lybrene tube method,researched the influence that different volumes of serum and LIM on red blood cell agglutination by the orthogonal principle, and determined the optimum parameters of shock through the observations. Simultaneously, the sensitivities of microplate method and Polybrene tube method were compared. Results The specifity of polybrene microplate method is 96.87% and its sensitivity is 98.44% by using the optimum reaction system,that centrifuging 1000 r/min, shocking 10 s,50 μL serum, 150μL LIM. Conclusion We developed a novel antibody screening method,which can instead of the traditional Polybrene tube method,it also could be batch operated and beautomatedeasily.
