摘  要:通过随机挑取克隆测序的方法从蜡梅花cDNA文库中获得了蜡梅谷氧还蛋白基因,命名为CpGRX1(GenBank登录号:JQ990126).该基因cDNA全长为491bp,其中包含的开放阅读框长为321bp,编码蛋白分子量约为11.18kD.生物信息学分析表明:CpGRX1蛋白属于典型的亚类I(即CPYC型)谷氧还蛋白.实时荧光定量PCR分析表明:CpGRX1基因在蜡梅的根、茎、叶、花等组织中均有表达,其在茎中的表达量显著高于其它组织;在开花过程中,CpGRX1基因在蕾期、盛开期和衰败期表达量较高,表明该基因可能与花的衰老过程相关.A glutaredoxin gene, named CpGRX1 (GenBank: JQ990126), was cloned by randomly selecting and sequencing a cDNA library constructed from Chimonanthus praecox. The cDNA sequence was 491 bp in full length and the open reading frame consisted of 321 bases encoding a protein with an estimated molecular weight of 11.18 kD. Bioinformatic analysis showed that the CpGRX1 protein belongs to Subclass I of glutaredoxin, i.e. CPYC type. According to real-time quantitative analysis, CpGRX1 gene is expressed in root, stem, leaf, flower and other tissues in C. praecox, the expression in stem being significantly higher than that in other tissues. At the flower-bud, bloom and withering stages, the expression of CpGRX1 gene was higher than that at other stages, which suggested that the gene might play an important role in flower senescence of C. praecox.
