摘  要:根据植物多酚氧化酶基因保守区域设计简并引物,采用同源克隆的方法,从花魔芋幼叶中首次克隆得到魔芋PPO基因的部分gDNA和cDNA片段.该gDNA和cDNA片段长度均为762bp,编码254个氨基酸残基.生物信息学分析表明具有酪氨酸酶家族的特征,具有保守的CuA和CuB结合区.系统进化树表明:花魔芋PPO蛋白与禾本科植物(大麦、小麦、玉米)最先聚为一类,亲缘关系最近.Polyphenol oxidase (PPO) is the enzyme responsible for enzymatic browning in Amorphophal- lus, which decreases the appearance quality and the commercial value of Amorphophallus products. In this research, degenerate primers were designed according to the conserved region sequences of previous published polyphenol oxidase genes (PPO), and the gDNA and cDNA fragment of the polyphenol oxidase gene in the young leaves of A. konjac was cloned for the first time, using the homology-based cloning method. The nucleotide sequence of the DNA and cDNA fragment was shown to be 762 bp in length and it encoded a protein of 254 amino acids. Bioinformatics analysis showed that PPO gene of Amorphophallus shared the characteristics of the tyrosinase family, having the conserved CuA and CuB binding regions. The phylogenetic tree showed that, phylogenetically, PPO protein of A. konjac was closely related to Gramineae plants (barley, wheat and maize), and they were clustered into one group.
