摘　　要：目的:确定血红素氧合酶同工酶1(HO-1)对银屑病患者外周血单一核细胞(PBMC)TNF-α和IL-10分泌的影响。方法:分离培养PBMC,应用免疫印迹法检测PBMC中HO-1的表达,用ELISA法检测上清中TNF-α和IL-10水平。结果:CoPP(HO-1特异性诱导剂-钴原卟啉)可诱导健康者和银屑病患者PBMC表达HO-1蛋白,表达量随CoPP浓度的增加而增加。4种不同浓度CoPP所诱导的HO-1均可抑制TNF-α的分泌和增强IL-10的分泌,其抑制和增强作用均在CoPP 10μmol/L浓度下达高峰。结论:HO-1可调节银屑病患者PBMC TNF-α和IL-10的分泌,从而可能对银屑病起到抗炎和免疫调节作用。Objective： To investigate the effects of hemeoxygenase- 1 （ H0-1 ） on the production of TNF- α and IL-10 by peripheral blood mononuclear cell （PBMC） in the patients with psoriasis vulgaris. Methods： PBMCs were obtained and cultured. The expression of H0-1 protein was tested by Western-blot. The level of TNF-α and IL-10 in the supernatants of cultured PBMC was measured by a specific enzyme-linked immu- nosorbent assay. Results： CoPP induced the expression of H0-1 protein in PBMC of both patients and con- trois in a dose-dependent manner. The secretion of HO-1 protein induced by different concentration of CoPP inhibited the secretion levels of TNF-α and enhanced the secretion of IL- 10, and the maximum effective con- centration was 10 p.mol/L. Conclusion： H0-1 can be induced from the secretion of TNF-ct and IL-10 PB MC in patients with psoriasis vulgaris, thus it is possible that H0-1 may be played in a role of anti-inflamma tory and immunomodulatory function in psoriasis.
