摘 要:目的观察复合羟丁基壳聚糖纳米载体携载CD+133抗体温敏膜对人脐静脉内皮细胞(HUVEC)增殖、迁移及血液相容性的影响。方法将HUVEC接种于裸钴铬合金片膜(裸金属支架组)、雷帕霉素药物涂层钴铬合金片膜(雷帕霉素支架组)、复合羟丁基壳聚糖纳米载体携载CD+133抗体温敏膜钴铬合金片膜(抗温敏膜组)。培养24 h后,在倒置相差显微镜下观察细胞形态;以MTT比色法检测细胞增殖活性,划痕试验检测细胞的迁移能力,ELISA检测各组细胞上清液中内皮素-1(ET-1)、降钙素基因相关肽(CGRP)、前列环素代谢物6-酮-前列腺素F1α(PGF1α)水平。结果 HUVEC在抗温敏膜上生长良好,形态正常。与裸金属支架组、雷帕霉素支架组相比,在抗温敏膜组存活的HUVEC数目增高(P均<0.05)。HUVEC在抗温敏膜组上迁移率高于裸金属支架组、雷帕霉素支架组(P均<0.05)。与裸金属支架组、雷帕霉素支架组相比,抗温敏膜组细胞分泌的ET-1水平降低;CGRP、PGF1α水平升高(P均<0.05)。结论复合羟丁基壳聚糖纳米载体携载CD+133抗体温敏膜有助于HUVEC的增殖和迁移,对HUVEC的血液相容性良好。Objective To investigate the influence of hydroxybutyl-chitosan nanoparticles carries CD133+ anti-thermosensitive membrane on the proliferation,migration and hemocompatibility of human umbilical vein endothelial cells (HUVECs).Methods HUVECs were inoculated into bare cobalt-chromium alloy film (bare metal stent group),sirolimus-eluting cobaltchromium alloy film (sirolimus stent group) and hydroxybutyl-chitosan nanoparticles carries CD133+ anti-thermosensitive membrane cobalt-chromium alloy film (anti-thermosensitive membrane group).After being cultured for 24 hours,the cell morphology was observed by inverted phase-contrast microscope.The cell proliferation was measured by MTT colorimeter.The ability of cell migration was detected by scratch tests.Enzyme linked immunosorbent assay (ELISA) was used to measure the levels of endothelin-1 (ET-1),calcitonin gene-related peptide (CGRP) and prostacyclin metabolite 6-keto-prostaglandin F1α (PGF1 α) in all groups.Results HUVECs grew well in the anti-thermosensitive membrane group,with normal morphology.Compared withthe baremetal stent and sirolimus stent groups,the number of survived HUVECs in the anti-thermosensitive membrane group was higher (all P <0.05).The migration ratio in the anti-thermosensitive membrane group was higher than that in the bare metal stent and sirolimus stent groups,respectively (all P < 0.05).Compared with bare metal stent and sirolimus stent groups,the concentration of ET-1 of anti-thermosensitive membrane group decreased,and increased in the CGRP and PGF1α levels (all P <0.05).Conclusions Hydroxybutyl-chitosan nanoparticles carries CD133+ anti-thermosensitive membrane cobalt-chromium alloy film can improvethe proliferation,migration and hemocompatibility of HUVECs.
