摘 要:目的研究都梁滴丸对硝酸甘油致偏头痛大鼠行为学及血浆一氧化氮(NO)、一氧化氮合酶(NOS)和降钙素基因相关肽(CGRP)水平的影响。方法 60只Sprague Dawley大鼠,雌雄各半,随机分为空白对照组、模型组、阳性对照组及都梁滴丸高、中、低剂量组,每组10只。除空白对照组外,其余各组大鼠采用硝酸甘油皮下注射制作偏头痛模型。各组大鼠灌胃给药后,分别观察其挠头次数,分光光度法和放射免疫法测血浆NO、NOS和CGRP水平。结果造模后30 min各组大鼠前肢挠头次数较空白对照组均显著增加(P<0.01)。与模型组比较,造模后60 min阳性对照组和都梁丸高剂量组大鼠挠头次数均显著减少(P<0.05);造模后90、120、150 min,都梁滴丸高、中、低剂量组和阳性组大鼠挠头次数均显著减少(P<0.05,P<0.01)。与空白对照组比较,模型组大鼠血浆NO、NOS、CGPR水平显著升高(P<0.01);与模型组比较,阳性组和都梁滴丸高、中、低剂量组NO、NOS、CGRP水平均显著下降,差异均有统计学意义(P<0.05,P<0.01)。结论都梁滴丸可通过降低血浆中NO、NOS、CGRP水平来发挥对偏头痛的治疗作用。Objective To observe the effect of Duliangdiwan on behavioral symptoms and levels of nitric oxide(NO), nitric oxide synthase(NOS) and calcitonin gene related peptide(CGRP) in blood plasma in the rat models of migraine induced by nitroglycerin. Methods Sixty Sprague Dawley rats were randomly divided into blank control group, model group, positive control group, Duliangdiwan high, middle and low dose group, with ten rats in each group. Except the blank control group, the rats in other groups were injected nitroglycerin to establish the migraine model. The frequency of scratching head was observed after intragastric administration drugs. The contents of NO, NOS and CGRP in plasma were measured by spectrophotometric method or radio-immunity method. Results The frequency of scratching head of rats in blank control group was significantly lower than those in other groups at 30 minutes after modeling( P 〈 0.01 ). Compared with model group, the frequency of scratc- hing head of rats in positive control group and Duliangdiwan high dose group were decreased(P 〈 0.05 ,P 〈 0.01 ) at 90,120, 150 minutes after modeling. The contents of NO, NOS and CGRP in plasma in model group were significantly higher than those in blank control group ( P 〈 0.05 ). Compared with model group, the contents of NO, NOS and CGRP in plasma in Duliangdiwan high, middle and low dose group decreased ( P 〈 0.05, P 〈 0.01 ). Conclusion Duliangdiwan can treat the migraine by de- creasing the contents of NO, NOS, CGRP in plasma.
