摘 要:目的:建立反相高效液相色谱法测定柴葛解肌汤中葛根素的含量。方法:采用Hypersil BDS C_(18)色谱柱,以乙腈-0.1%磷酸水溶液为流动相梯度洗脱。流速为1.0 ml·min^(-1),检测波长250 nm,柱温30℃。结果:葛根素进样量在0.032 3~0.323 2μg范围内呈良好的线性关系(r=0.999 8,n=6);平均加样回收率为101.4%,RSD为2.9%(n=6)。结论:该法具有简便易行、检测灵敏、重复性好等优点,可用于柴葛解肌汤中葛根素的含量测定。Objective: To establish a method for the determination of Puerarin in Chaige Jieji decoctions by HPLC. Methods: The separation was carried on a Hypersil BDS C18 column with acetonitrile-0.1% phosphoric acid as the mobile phase with gradient elution, and the column temperature was maintained at 30℃. The detecting wavelength was 250 nm, and the flow rate was 1.0 ml ·min^-1 Results: The linearity of the calibration curve was good within the range of 0.032 3-0.323 2μg for puerarin. The average reeoverywas 101.4% (RSD = 2.9% ,n = 6). Conclusion: The method can sensitively detect puerarin with a simple pretreatment, and has a broad linear range with good repeatability, which can be used in the determination of puerarin in Chaige Jieji decoctions.
