摘  要:目的探讨富含Trp/Arg六肽对糖尿病小鼠皮肤创伤愈合的影响。方法经小鼠腹腔注射链脲佐菌素建立小鼠糖尿病模型,在糖尿病小鼠背部切取直径8 mm的圆形全层皮肤,形成皮肤损伤模型。将皮肤损伤模型小鼠随机分为富含Trp/Arg六肽RRWWCR、RRWWFR、RRWWTR组和生理盐水组,分别在创伤形成同时,在伤口处擦拭0.1 mg/ml相应药物,每只0.2 ml,开放式治疗10 d,每天1次。每天观察创面形态变化,并测量创面直径;每5 d取附带周围正常组织的创面全层皮肤,进行组织病理学观察。结果 RRWWCR和RRWWTR可促进创面组织的修复,表现为加强早期炎症反应、促进新生血管、肉芽组织和再上皮化形成以及缩短创面愈合时间。结论富含Trp/Arg六肽对糖尿病小鼠皮肤创伤愈合具有促进作用,其第5位氨基酸残基极性可极大地影响其促修复能力。Objective To investigate the effect of Trp / Arg-rich hexapeptides on skin wound healing in diabetic mice. Methods Diabetic mouse model was established by intraperitoneal injection with streptozotocin. Full-thickness circular skin at a diameter of 8 mm was removed from the back of each model mice. The mice with skin wound were divided into four groups,15 for each,of which the wounds were smeared with 0. 2 ml(0. 1 mg / ml) of RRWWCR,RRWWFR, RRWWTR and physiological saline as control respectively,once a day for 10 d. The wound surface was observed for morphological change and determined for diameter daily. The full-thickness skin of wound surface,with normal skin tissue around,was collected every 5 d and observed for histopathology. Results RRWWCR and RRWWTR promoted the skin wound closure of diabetic mice,expressed as enhancing the initial inflammatory reaction,promoting the formations of angiogenesis and granulation tissues as well as re-epithelialization, and shortening the time for wound healing. Conclusion Trp / Arg-rich hexapeptides promoted the skin wound healing of diabetic mice,of which the polarity of the fifth amino acid residue extremely influence the promoting effect on tissue repair.
