摘　　要：那格列奈是一种口服非磺脲类短效胰岛素分泌促进剂,是临床用于治疗2型糖尿病的常用药物之一。近年的研究发现,其降糖作用机制还包括促进胰高血糖素样肽1(GLP-1)的释放、抑制二肽基肽酶Ⅳ(DPP-Ⅳ)和胃促生长素活性。那格列奈的晶型、结构改造以及剂型变化对其药效学和药动学有一定的影响。无论是单独用药还是与其他降糖药物及心血管药物联用,那格列奈均可发挥稳定的临床疗效,且可与其他药物发挥协同作用,不良反应少。Nateglinide, an oral non-sulfonylurea short-acting insulin secretion promoter, is one of the commonly used drugs for treating type 2 diabetes mellitus. Recent studies suggest that promoting the release of GLP- 1, inhibiting the activity of DPP- IV and ghrelin may also be associated with its hypoglycemic activity. The changes of its polymorphs, structure and formulations transformation may impact on the efficacy as well as its pharmacokinetic characteristics. Nateglinide could play a stable clinical efficacy either alone or in conjunction with other antidiabetic and cardiovascular drugs. It can bring about synergies with other drugs with little adverse reactions.
