摘  要:目的:观察复方补骨脂搽剂对白癜风豚鼠模型外周血中CD4+/CD8+比值以及对化学脱色皮损区TNF-α、IL-6表达的影响,探讨复方补骨脂搽剂对促进实验动物黑色素形成的作用。方法:选取健康豚鼠60只随机分为正常组、模型组、复方补骨脂搽剂高、中、低剂量组和阳性药组(复方卡力孜然)共6组,除正常组外用化学脱色法建立白癜风皮损模型,对相应组别采用不同剂量的复方补骨脂搽剂或阳性药治疗。免疫组化检测各组皮损区TNF-α及IL-6表达水平,流式细胞仪检测动物外周血中CD4+和CD8+水平,计算CD4+/CD8+比值水平并进行分析比较。结果:模型组CD4+/CD8+比值显著低于正常对照组,TNF-α和IL-6表达明显高于对照组,差异具有统计学意义(P<0.01);复方补骨脂搽剂高、中剂量组和阳性药组TNF-α及IL-6表达与模型组相比明显下降,CD4+/CD8+比值显著升高,差异具有统计学意义(P<0.05)。结论:实验动物皮损区TNF-α及IL-6表达水平的上升可能参与了白癜风的发病,而复方补骨脂搽剂可明显降低TNF-α及IL-6的表达,并改善实验动物细胞免疫状况,进而对白癜风实现一定的治疗作用。OBJECTIVE To observe the effects of compound scurfpea fruit liniment on the peripheral blood CD4+ / CD8+ ra- tio and the expression of TNF-α, IL-6 in the skin of chemical decolorization area in guinea pig model of vitiligo, to explore the effect of compound scurfpea fruit liniment on promoting the formation of melanin in experimental animals, METHODS 60 Healthy guinea pigs were randomly divided into 6 groups: normal group, model group, compound scurfpea fruit liniment high, medium and low dose group and positive medicine group, the vitiligo model was established with chemical decolorization of the skin method,and treated with different doses of compound scurfpea fruit liniments or positive medicine. Immunohistochemical method was used to detect the TNF-A and IL-6 expression levels in the lesion area. RESULTS The content of CD4+ and CD4+/ CD8* ratio in model group was significantly lower than that in normal control group, the TNF-α and IL-6 expression was significantly higher than the control group, the difference was statistically significant (P〈0. 01 ). Compound scurfpea fruit liniment high, middle dose group and positive medicine group decreased TNF-α and IL-6 expression significantly as compared with model group, and significantly increased the CD4+ / CD8+ ratio with statistically significant difference (P〈0. 05). CON- CLUSION TNF-α and IL-6 expression level rise in experimental animal skin lesion area may be involved in the pathogenesis of vitiligo, compound scurfpea fruit liniment can obviously reduce the expression of TNF-α and IL-6, and improve the experimental animal cell immune status, so it has a certain therapeutic effect on vitiligo.
