可溶性Ⅰ型胶原蛋白对移植肝细胞凋亡的抑制作用

摘　　要：目的　研究可溶性Ⅰ型胶原蛋白对自体移植肝细胞凋亡的抑制作用及相关机制。方法　建立自体肝细胞脾内移植大鼠模型 ,按肝细胞处理的不同分为肝细胞悬液不处理组 (A组 )、拮抗型抗整合素 β1单克隆抗体及Ⅰ型胶原蛋白处理组 (B组 )以及Ⅰ型胶原蛋白处理组 (C组 ) ,应用99mTc AnnexinV的单光子发射计算机体层摄影术 (SPECT)显像测定肝化脾对99mTc AnnexinV的摄取率 ,采用原位末端标记法 (TUNEL法 )检测肝细胞凋亡指数 ,并用免疫组织化学法检测移植肝细胞磷酸化酪氨酸蛋白的表达。结果　C组的移植肝细胞凋亡指数和99mTc AnnexinV摄取率分别为 (3 .86±1.37) %和 5 5 1.2± 98.3,均显著低于A组和B组 (P <0 .0 1) ,其磷酸化酪氨酸蛋白表达强度在 + +以上者为 75 % ,高于A组和B组 (P <0 .0 5 ) ,其磷酸化酪氨酸蛋白的表达与凋亡指数呈负相关。Objective To study the inhibitory effect of soluble collagen I on autospleen transplanted hepatocytes and its mechanism.Methods Scintigraphic imaging studies with 99m Tc Annexin V was performed , as well as TUNEL method was used to evaluate the apoptosis inhibitory effect of soluble collagen I on autospleen transplanted hepatocytes. In addition, immunohistochemical study was adopted to assess the expression of phosphotyrosine proteins of transplanted hepatocytes treated with soluble collagen I. Results The group treated with soluble collagen I showed a 99m Tc Annexin V uptake rate of hepatocized spleen as 551.2 ± 98.3 , and an apoptotic index of transplanted hepatocytes as (3.86 ± 1.37)?% , obviously lower than those in the control group and the group pre treated with antagonistic integrin β1 anibodies ( P < 0.0l ). Besides, its“++' positive rate of phosphotyrosine proteins in the group treated with soluble collagen I was higher than in the other 2 groups ( P < 0.05 ). The expression of phosphotyrosine proteins was negatively related to apoptotic index of transplanted hepatocytes. Conclusions Soluble collagen I could inhibit apoptosis of transplanted hepatocytes by way of interacting with integrin β1 on cell surface and phosphorylating tyrosine proteins.
