摘 要:目的观察高湿环境对SD大鼠肾功能的影响,探讨水通道蛋白-2(AQP2)和加压素受体-2(V2R)在高湿环境大鼠肾脏中的表达及其意义。方法 20只SD大鼠随机分为对照组和高湿组,每组10只。高湿组大鼠每日放入人工气候箱10 h[温度25℃,相对湿度(RH)为(90±2)%],对照组置常温常湿环境[温度(25±2)℃,RH(55±5)%]。处理20 d后,荧光定量PCR检测肾髓质AQP2和V2R mRNA表达水平,Western blot法检测肾髓质AQP2、磷酸化AQP2(p-AQP2)以及V2R的蛋白表达水平。结果与对照组相比,高湿组大鼠血尿素氮(BUN)、血肌酐(SCr)显著升高(P<0.05);肾髓质AQP2和V2R mRNA表达较对照组显著降低(P<0.01),AQP2、V2R的蛋白表达水平和AQP2的磷酸化水平较对照组显著降低(P<0.05)。结论高湿环境可对大鼠肾功能造成一定损害,且使V2R和AQP2的表达水平降低,AQP2磷酸化水平也降低,提示V2R和AQP2可能参与了高湿环境引起的肾功能损害的发生发展。Objective To observe the influence of high humid environment on SD rats'renal function, and to discuss the expression and significance of aquaporin protein-2 ( AQP2 ) and vasopressin receptor-2 ( V2R ) in rats ' kidneys in high humidity environment. Methods Twenty SD rats were randomly divided into control and high humidity groups with l0 ones in each group. The rats in the high humidity group were carried daily into artificial climatic cabinet for 10 hours [ the temperature was 25 ℃, and the relative humidity was (90 ± 2)% ]. The rats in the control group were put in the environment with common temperature [ the temperature was (25 ± 2 )℃, and the RH was (55 ± 5 )% ]. After 20 d ,fluorescent quantitation PCR was used to detect the mRNA expression levels of renal medull AQP2 and V2,and Western blotting method was used to detect the protein expression levels of AQP2, phosphotylated AQP2 (phosphorylation of aquaporin protein-2, p-AQP2) , and V2R. Results Compared with control group, the blood urea nitrogen (BUN) and serum creatinine(SCr) in the high humidity group significantly increased compared with those indexes in the control group (P 〈 0.05). The mRNA expression levels of renal medulla AQP2 and V2R and the protein expression levels of AQP2, p-AQP2, and V2R in the high humidity group all significantly decreased compared with those levels in the control group [ ( P 〈 0.01 ) and ( P 〈 0.05 ) 1. Conclusion High humidity environment can cause damage of the renal function of the rats and decrease the expression levels of V2R and AQP2 and the phosphorylation level of AQP2 ,which indicates that V2R and AQP2 may be involved in the development of renal function lesion caused by the humid environment.
