摘 要:目的:探讨米诺环素(minocycline)对大鼠脊髓损伤(SCI)早期的保护作用和机制.方法:108只SD大鼠随机分为3组:对照组(A组=36只)、损伤组(B组=36只)和损伤后米诺环素治疗组(C组=36只),采用改良Allen's打击法致伤B组和C组大鼠T8~T11脊髓,对照组不损伤脊髓.治疗组于术后1h腹腔注射米诺环素(90mg/kg),之后每隔12h给药1次,对照组和损伤组在相同时点腹腔注射相同体积生理盐水.B组和C组于术后2h、6h、12h、24h、48h、72h处死大鼠取损伤段脊髓标本,A组在相应时间点取相应节段脊髓标本,切片后行HE染色组织病理学检查和免疫组织化学检测TNF-α表达.结果:脊髓损伤早期损伤脊髓取材免疫组化镜检后发现A组大鼠脊髓TNF-α呈弱阳性表达;B组和C组伤后2h即有TNF-α表达,12h达到高峰,之后逐渐下降;B组伤后72h TNF-α表达仍较A组高(P<0.05),而C组在伤后72h即恢复到对照组水平(P>0.05),C组各时间点TNF-α表达均较损伤组低(P<0.05).结论:米诺环素可显著降低脊髓损伤早期组织中TNF-α的表达,为其应用于临床治疗急性脊髓损伤提供实验依据.Objective: To investigate the effect of Ininocycline on the expression of tumor necrosis factor-α (TNF-α) in acute spinal cord injury, and the mechanism ol mmocyclme on spinal cora injury in. Method: 108 rats were randomly divided into control group, injury group and the minocycline--intervention group with 36 rats in each group. Allen's method was used to induce spinal cord injury both in injury group and in intervention group. The spinal cords of rats in control group are not injured. In 1 hours after the operation, the rats in the treatment group were injected with minocycline(90mg/kg), and given medicine every 12 hours. The injured spinal cord segments were collected and processed pathologically at 2h, 6h, 12h, 24h, 48h and 72h after operation. At the same time point, control the corresponding segment of spinal cord samples group rats was removed. After slicing, tissues were stained with HE examination and immunohistochemical detection of TNF-α. Results: The expressions of spinal cord injury at TNF-- a of rates in A is weakly positive. The ex- pression of TNF-α increased at 2 hours after injury in B group and C, and reached the peak atl2 hours, then decreased gradually. The expression of TNF-α of tissue in injury group was higher than that in control group (P 〈 0.05) in 72h after injury. The expression of TNF-α in intervention group returned to the level of the control group (P 〉 0. 05) in 72h after injury. TNF-α expression in intervention group was lighter than that in injury group at each time point (P 〈0.05). Conclusion :Minocycline could significantly reduce the expression of TNF-α in SCI rats and provide experimental basis for the treatment of spinal cord iniury.
