摘 要:目的:研究骨髓单个细胞移植脊髓半横断大鼠脊髓损伤区域后髓鞘碱性蛋白(myelin basic protein,MBP)表达的影响。方法:取Wistar大鼠胫骨及股骨提取骨髓单个核细胞,采用密度梯度离心法分离骨髓单个核细胞,制成细胞悬液。建立大鼠脊髓右半横断模型,造模后分为假手术组、模型组、骨髓单个核细胞移植组。于模型建立成功后7d,14d,21d,28d手术的大鼠进行BBB评分从而评估损伤脊髓神经功能恢复情况,利用RT-PCR观察MBP mRNA的表达变化。结果:术后各时间点假手术组BBB评分均明显高于骨髓单个核细胞移植组、模型组(P<0.05);术后随时间延长骨髓单个核细胞移植组、模型组BBB评分逐渐增加,14、21、28d时骨髓单个核细胞移植组BBB评分明显优于模型组(P<0.05)。PT-PCR测试各时间点骨髓单个核细胞移植组和模型组大鼠脊髓中MBP mRNA的表达量高于假手术组,且骨髓单个核细胞移植组高于模型组(P<0.05)。结论:骨髓单个细胞移植可提高脊髓组织中MBP的表达,有助于大鼠半横断脊髓损伤神经功能的恢复。Objective: To study the expression of Myelin Basic Protein (MBP) in spinal cord after trans- plantation of bone marrow mononuclear cell transplantation in spinal cord hemi- section injury rat models. Methods: Bone marrow was extracted from tibia and femur of wister rats. Bone marrow mononuclear cells (BMMNCs) were isolated by density gradient centrifugation and cell suspension was prepared. After establish- ment of spinal cord hemi--section injury rat models, rats were divided into sham surgery group, control group and bone marrow mononuclear cells transplantation group. In 7th day, 14th day, 21th day and 28th day post- surgery and rat models were assessed for locomotor functional recovery using BBB score and change in the ex- pression of MBP mRNA by RT--PCR. Results: At each time point after surgery, BBB scores in sham surgery group were significantly higher than those in bone marrow mononuclear cells transplantation group and control group (P 〈 0. 05). At subsequent time period, there was gradual increase in BBB scores in both bone marrow mononuclear cells transplantation group and control group. At 14th day, 21th day and 28th day post--surgery, BBB scores in bone marrow mononuclear cells transplantation group were significantly higher than those in the control group (P 〈 0. 05). Expression of MBP mRNA at each time point in spinal cord were assessed by RT-- PCR and compared between sham surgery group, bone marrow mononuclear cells transplantation group and control group. Level of MBP mRNA expression were higher in bone marrow mononuclear cells transplantation group compared with those in control group (P 〈 0.05). Conclusion: Bone marrow mononuclear cells trans- plantation accentuates the expression of MBP in spinal cord tissue and thus helps the recovery of neurological function in rat models with spinal cord hemi -- spinal cord injury.
