摘　　要：【目的】以地高辛 (DIG)标记探针检测汉族肥胖者瘦素 (leptin)基因的表达。【方法】以DIG用随机引物法标记瘦素cDNA片段 ,以此探针 ,采用斑点杂交方法检测 11例汉族正常体重者 (体重指数 ,MBI为 2 1 0kg/m2 ± 1 5kg/m2 )和 12例汉族肥胖者 (MBI为 2 8 5kg/m2 ± 2 3kg/m2 )腹部皮下脂肪组织瘦素mRNA的表达水平。【结果】DIGleptincDNA探针可检出 5pg的 pUC leptinDNA ,与pUC19DNA无杂交信号。肥胖组腹部皮下脂肪组织瘦素mRNA相对含量明显高于对照组 (312 8± 10 8 9vs175 9± 81 5 ,P <0 0 5 ) ;且与BMI呈正相关 (r =0 5 6 ,P <0 0 5 )。【结论】肥胖者脂肪组织瘦素mR NA水平明显增高 ,且与BMI呈正相关。Objective To study the expression of leptin mRNA in Chinese obese subjects using dot blot hybridization with a digoxigenin labeled probe. Methods 11 Chinese nonobese subjects (bodymass index, BMI 21.0 kg/m 2±1.5 kg/m 2) and 12 Chinese obese subjects(BMI 28.5 kg/m 2±2.3 kg/m 2) participated in the study. The human leptin cDNA probe was labeled wih digoxigenin(DIG) by the random priming method. Expression of leptin mRNA in abdominal adipose tissue has been examined using dot blot hybridization with this probe. Results The expression of leptin mRNA was significantly higher in obese subjects than in non-obese ones (312.8±108.9 vs 175.9±81.5, relative units, P<0.05), and correlated with BMI (r=0.56, P<0.05). Conclusions Leptin mRNA level is high in obese subjects, and correlated with BMI.
