摘 要:目的观察补阳还五汤中不同黄芪剂量对大鼠大脑皮质神经细胞缺血缺氧再灌注损伤的影响。方法将新生sD大鼠大脑皮质细胞取出分成8组,第1组为正常细胞培养组;第2组为大鼠皮质神经细胞缺氧缺糖再给氧损伤(模拟神经细胞缺血缺氧再灌注损伤)造模组,第3至8组在造模的基础上,分别加入黄蓖剂量依次为0、30、60、90、120、150g的补阳还五汤黄芪剂量加减方的含药血清。双染法荧光显微镜观察细胞的形态变化和坏死、凋亡的发生率,流式细胞仪测定细胞凋亡情况,评价含药血清的影响。结果黄芪剂量在0~60g时,增加其剂量则细胞死亡率、凋亡率均明显下降(P均〈0.05);黄芪剂量增加至90、120、150g后,细胞死亡率下降明显(P均〈0.05),但该i组间细胞死亡率相近(P〉0.05);细胞凋亡率在60g后不再下降,至90、120、150gi组间稳定在同一水平(P〉0.05)。结论补阳还五汤中黄芪剂量在较小时逐渐增加可能提高对缺血缺氧再灌注损伤细胞坏死和凋亡的抑制作用,而当黄芪剂量增加到90g后。再增加剂量可能并不能提高其抑制凋亡的作用。Objective To observe the effect of different doses of Astragalus in Buyanghuanwu decoction (BYHWD) on hypoxia-ischemia-reperfusion injury of cerebral cortical neurons in rats. Methods The cerebral cortex cells removed from newborn SD rats of were divided into eight groups: normal control group; model group (establishing the model of hypoxia-hypoglycemia-reoxygenation injury) ; 3 to 8 drug groups: on the basis of molding the serum containing different doses of Astragalus (0,30,60,90,120,150g) in modified BYHWD were respectively added in the cortical cell culture solution; the serum of no drug in normal control group and model group was also added in the cortical cell culture solution. Using double staining method, the cell morphological change, necrosis and apoptosis incidence were observed under florescence microscope, and cell apoptosis was detected by flow cytometer alter incubation for four hours. The influence of serum containing drug on cultured cells was assessed. Results When the doses of Astragalus were 0 to 60g, both the cells mortality and apoptosis rate decreased with the increase of dose; when the doses increased to 90,120 and 150g,the cell mortality decreased more significantly (all P 〈 0.05), but there was no significant difference among the three groups (P 〉 0.05 ) ; while after the doses increased to 60g, the cell apoptosis rate did not further decrease instead of same levels when being 90,120 and 150g (P 〉 0.05 ). Conclusions When the Astragalus dose in modified BYHWD is smaller, increasing its doses might increase the inhibiting effect against the cell necrosis and apoptosis induced by hypoxia-ischemia-reperfusion injury, but after increasing to 90g,the further increase of Astragalus doses could not enhance the effect of inhibiting cell apoptosis.
