摘 要:目的研究浒苔多糖(EP)的分离纯化条件和结构特点。方法以福建浒苔为研究对象,采用二乙氨基乙基纤维素(DEAE纤维素)和Sephadex-100凝胶分离纯化浒苔多糖,用红外线光谱分析结构,用气相色谱分析单糖组成。结果浒苔多糖经DEAE纤维素分离得到4个组分,EP1、EP2、EP3和EP4,其中EP2在所回收的多糖中所占的比例最大,达到81%。EP2经Sephadex-100分离时,时呈现单一对称峰;红外线光谱分析显示,EP2为糖类化合物,且有硫酸酯结构;经气相色谱检测显示,EP2由鼠李糖、葡萄糖醛酸、阿拉伯糖、岩藻糖、甘露糖和半乳糖组成,各种单糖的比例为5.12∶1.32∶3.38∶1.62∶1∶1.03。结论 EP的主成分为EP2,EP2是一种含有硫酸酯的复合多糖。Objective To study the separation and purification condition and structure features of polysaccharide from Entero- morpha. Methods The Enteromorpha produced in Fujian province was taken as the research object. DEAE - cellitlose and Sephadex - 100 were used to separate and purify the polysaccharide from Enteromorpha(EP). Infrared spectrum was used to an- alyze the structure of EP and gas chromatography was used to study the composition of monosaccharide of EP. Results Four components were separated from EP by DEAE - cellulose, they were EP - 1, EP - 2, EP - 3, EP - 4. EP2 accounted for the largest proportion of the recycled polysaccharide, reaching 81%. EP -2 showed a single symmetric peak when separated by Sephadex - 100 ; Infrared spectrum showed that EP - 2 was a kind of carbohydrate with thiol esters. GS showed that EP - 2 was made up of rhamnose, glucuronic acid, arabinose, fueose, mannose and galaetose, and the proportion of each monosaccharide was 5.12: 1.32: 3.38: 1.62: 1: 1.03. Condusion The major components of EP is EP - 2. EP - 2 is a kind of polysaccharide containing thiol esters.
