摘 要:目的观察续命汤对局灶性脑缺血中风大鼠血清白介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)含量的影响。方法将清洁级SD大鼠分为分假手术组、模型组、尼莫地平组及续命汤低、中、高剂量组6组,通过栓线法制备局灶性脑缺血中风大鼠模型,观察并记录SD大鼠造模后神经功能评分,测定血清IL-1β和TNF-α含量。结果造模后大鼠神经行为学评分显著升高,IL-1β和TNF-α含量增加,续命汤各剂量能拮抗以上作用,以续命汤中、高剂量为显著(P<0.05或P<0.01),其疗效与尼莫地平组相当(P>0.05)。结论续命汤能降低IL-1β、TNF-α的过度表达,减少缺血中心区和周围区神经元损伤,减轻脑缺血和水肿,进而实现保护血脑屏障的作用。Objective: To observe the effect of Xuming Decoction on interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) content of rat with focal cerebral ischemic stroke. Methods: The clean grade SD rats were di- vided into six groups: sham operation group,model group,nimodipine group,Xuming Decoction of low dose group, middle dose group, and high dose group. The model of focal cerebral ischemic stroke rats was built by using thread embolism method. Neurological score ,expression of IL-1β and TNF-α content in rat brain detected by im- munohistochemical method were observed and recorded after SD rats modeling. Results: The neurological score was significantly higher,the expression of IL-1β and TNF-α were increased after rats modeling while all Xuming Decoction treatment groups antagonize these effects ,in which the effects of middle dose group and high dose group were obvious. Curative effects of the two groups were equivalent to those of Nimodipine group. Conclusion: Xum- ing Decoction plays an important role in reducing the excessive release of IL-1β and TNF-α,preventing from cerebral ischemia and edema, and alleviating cerebral ischemia and edema to protect the blood-brain barrier.
