摘 要:目的:通过动物实验验证中药柴胡及各有效成分(挥发油、皂苷)的解热作用并揭示其作用机制。方法:向大鼠背部皮下注射干酵母混悬液制取发热模型,随机分为6组,每组10只,灌胃给药。正常对照组:生理盐水10 mL/kg;模型对照组:生理盐水10 mL/kg;阿司匹林组:0.1 g/kg;柴胡挥发油组:0.25 g/kg;柴胡皂苷组:0.35 g/kg;柴胡水煎液组:10 g/kg,给药容量均为10 mL/kg。测量致热后大鼠体温变化验证其解热作用;测量大鼠下丘脑和血浆中环磷酸腺苷(cAMP)含量以及脑腹中隔区和血浆中精氨酸加压素(AVP)含量。结果:中药柴胡及各有效成分(挥发油、皂苷)对皮下注射干酵母混悬液引发大鼠的发热反应均可起到降温作用;可降低致热大鼠下丘脑中cAMP及脑腹中隔区AVP含量,升高血浆AVP含量,对血浆cAMP含量无显著影响。结论:中药柴胡及各有效成分(挥发油、皂苷)均具有明显的解热作用,作用机制与体内cAMP及AVP的合成分泌有关。Objective:To testify the function of relieving the fever of ChaiHu (Bupleurum chinense) and its active ingredients(volatile oil and saponin) and reveal its meachanism. Methods:The rat models were established by subcutaneous injection of dried yeast suspension and randomized into six groups, ten rats each group, intragastric administration. Normal control group:physiological saline 10 mL/kg;model control group:physiological saline 10 mL/kg;aspirin group:0.1g/kg;volatile oil group of ChaiHu:0.25 g/kg;saponin group of ChaiHu:0.35 g/kg;ChaiHu decoction group:10 g/kg, the volume was 10 mL/kg. The changes of body temperatures of the rats were measured in order to testify its function of relieving fever;the contents of cyclic adenosine monophosphate (Camp) in the blood plasma and hypothalamus of the rats were detected as well as the contents of arginine vasopressin(AVP) of the blood plasma and ventral septal areas to reveal the mechanism of Chaihu relieving fever. Results: ChaiHu and its active ingredients (volatile oil and saponin) could lower the temperature in the fever of rats caused by subcutaneous injec-tion of dried yeast suspension;it decreased the contents of AVP in the ventral septal areas and the contents of cAMP in the hypothalamus of rat model, raised the contents of AVP in the blood plasma and showed no influence on the contents of cAMP in the blood plasma. Conclusion:ChaiHu and its active ingredients(volatile oil and saponin) could effectively relieve the fever, its mechanism might be related to the synthesis and the secretion of cAMP and AVP in the body.
