摘 要:目的研究PPARγ基因单核苷酸多态性(SNPs)与妊娠期糖尿病(GDM)的关联。方法测定72例湖北汉族GDM孕妇(GDM组)及78名正常健康孕妇(对照组)PPARγ基因Pro12Ala(rs1801282)SNP多态性,分析其与妊娠期糖尿病(GDM)的关系。基因分型采用聚合酶链式反应-限制性片段长度多态性方法。结果 GDM组PPARγ等位基因频率明显高于对照组(P<0.05);与对照组比较,GDM组患者2 h血糖、空腹总胆固醇、空腹三酰甘油值明显升高,差异具有显著性(P<0.05)。GDM组不同基因型人群,PPARγ两种基因型组间血糖和胰岛素水平差异无显著性(P>0.05)。结论 PPARγ基因多态性与湖北汉族孕妇妊娠期糖尿病无直接关系,但可能参与GDM的血糖水平和脂质代谢的调节。Objective To research PPARγ gene single nucleotide polymorphisms (SNPs) associated with gestational diabetes. Methods PPAR,y gene Prol2Ala rs1801282 SNP polymorphism were determined in 72 cases of GDM pregnant women ( GDM group ) and 78 normal healthy pregnant women( normal healthy group). The relations with gestational diabetes mellitus (GDM) were analyzed. The method was Genoty- ping by using a polymerase chain reaction restriction fragment length polymorphism ( PCR - RFLP). Results PPAR gamma allele frequency GDM group was obviously higher than that of control group ( P 〈 0.05 ). Rs12636454AA genotype 3 acyl glycerin (TG) levels higher than that of AB+ BB genotype ( P 〈 0.05 ). Compared with control group, patients with GDM group 2 h blood glucose, fasting total cholesterol, fasting three acyl glycerin value increased significantly( P 〈0.05 ). GDM group of people of different genotypes and PPAR gamma two genotypes group there was no significant difference between blood sugar and insulin levels ( P 〉 0.05 ). Conclusion PPARγ gene and hubei han Chinese pregnant women gestational diabetes has no direct relation, but the level of the blood sugar may participate in GDM and the regulation of lipid metabolism.
