摘 要:建立HPLC法测定沙棘叶、果实、枝条中齐墩果酸和熊果酸含量。在色谱柱(岛津ODS柱4.6mm×250mm,5μm),流动相(甲醇-0.2%磷酸溶液(87∶13)),检测波长210nm,柱温30℃,流速1.0mL/min条件下进行测定。结果显示:齐墩果酸进样量在0.7-2.1μg范围内,峰面积与进样量之间线性关系良好,平均回收率为99.88%;熊果酸进样量在0.75-2.25μg范围内,峰面积与进样量呈良好线性关系,平均回收率为100.8%。本方法可测定沙棘叶、果实、枝条中齐墩果酸和熊果酸的含量,结果准确,重复性好。To set up an RP-HPLC method of simultaneous determ-ination for content analysis of olea-nolic acid and ursolic acid in the leaves,fruits and branches of Hippophae rhamnoides. Method:the op-timum HPLC condition was as follows: Shimadzu ODS col-umn (4.6mm×250mm, 5μm), mobile phase:methanol-0.2% Phosphoric acid(87:13),detection wavelength: 210nm, column temperature: 30℃,flow rate:1.0mL/min. Results: there is a good linearity relationship between peak areas and injectionquantity for both. The average recovery was 99.88%, under the injectionquantity of oleanolic acid in the range of 0.7-2.1μg and average recovery was 100.8% for ursolic acid ranging from 0.75 to 2.25μg..Conclu-sion: The method could detect oleanolic acid and ursolic acid in the leaves, fruits and branches of Hip-pophae rhamnoides simultaneously. The HPLC measurement was reasonable, stable and accurate.
