摘  要:目的 研究鼠缺血再灌注(I/R)心肌细胞肌浆网钙ATP酶(SR·Ca2+-ATPase,SERCA2a)的蛋白及其基因表达的变化,探讨基固调控在。肌保护中的作用。方法 将SD大鼠分为正常对照组与I/R组,利用离体心工作模型进行灌注,采用Western-Blot方法检测心肌细胞SERCA2a蛋白的含量,利用Northern-Blot方法测定SERCA2a mRNA相对含量。结果I/R心肌细胞的SERCA2a蛋白含量(0.538±0.006vs1.020±0.015)及其mRNA相对量(062±0.008vs0.95±0.009)均明显下降,分别下降34.7%与47.3%,其变化状态与其心肌功能变化一致。结抡 缺血再灌注过程严重影响了心肌细胞 SR·Ca2+-ATPase蛋白及其基因的表达,SERCA2a基因表达显著下降可能是心肌I/R损伤的重要机制;若采取有效措施进行基固调控,增强SERCA2a基因的表达,可能起到保护I/R心肌细胞、增强心肌功能的作用。Objective To observe the changes of sarcoplasmic reticulum Ca2+ - ATPase (SERCA2a) protein and gene expression in ischemic reperfusion (I/R) myocytes; To reveal the effects of gene regulation in myocardial protection. Methods Rats were divided into two groups (normal control group and I/R group) , An isolated rat heart working model was used, to determine the relative levels of SERCA2a protein and mRNA in different groups by Western Blot and Northern Blot analysis. Results The protein and mRNA levels declined markedly in the I/R group (0.538 ± 0.006 vs 1.020 ± 0.015 in protein, 0.62 ± 0.008vs 0.95 ± 0.009 in mRNA, P<0.01) for 34.7% and 47.3% respectively, The changes were correlated closely with myocardial function. Conclusion Ischemic reperfusion may adversely affect SERCA2a expression on protein and gene level, Depressed SERCA2a expression in I/R course may be an important mechanism in myocardial ischemic reperfusion injury. It may be helpful to protect myocytes from ischemic reperfusion injury and to improve myocardial function by upregulating gene expression.
