摘 要:研究马齿苋多糖(POP)对环境雌激素引起的生殖氧化损伤的影响,为其功能性食品开发提供参考。炔雌醇(EE2)处理组,按0.1 mg/kg、0.5 mg/kg、1 mg/kg、5 mg/kg大鼠腹腔注射EE2,POP对比组同时注射10%POP,测定大鼠睾丸相对质量、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)、丙二醛(MDA)和过氧化氢酶(CAT)水平,光镜观察睾丸组织结构。EE2处理组,注射1 mg/kg EE2后,睾丸组织结构紊乱,睾丸萎缩,相对质量下降(P<0.05);且EE2处理组,随EE2剂量增加和作用时间延长,T-AOC、CAT、SOD和GSH-Px水平降低(P<0.05),MDA水平升高(P<0.05)。POP对比组,组织结构未发生明显变化,T-AOC、CAT、SOD和GSH-Px水平稳定(P>0.05),个别组升高显著(P<0.05),MDA水平稳定(P>0.05)。提示炔雌醇具有生殖毒性,造成脂质过氧化,与ROS密切相关,POP具有抗生殖氧化损伤作用,维持ROS和抗氧化酶防御系统之间的平衡。The protection mechanism ofPortulace Oleracea polysaccharide (POP) on reproductive toxicity and oxidative damage induced by EEs in male rats was studied to provide the experimental basis for development and application of POE The male rats were established as fertility problems model by intraperitoneal injecting ethinyloestradiol (EE2), with the injecting dose of 0.1 mg/kg, 0.5 mg/kg, 1 mg/kg and 5 mg/kg, and meanwhile intraperitoneal injection of 10% POP as the comparative group.Absolute and relative testicular weights were measured. Morphological changes oftesticular were observed in the light microscope. The contents of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), total antioxide capacity (T-AOC), malondialdehyde (MDA) and catalase (CAT) in testicular tissue and plasma were detected. In EE2 processed groups, testicular weight decreased significantly (P〈0.05) and led to shrink when injection of lmg/kg EE2. The antioxidant enzymes including SOD, GSH-Px, CAT and T-AOC in testicular tissue all decreased (P〈0.05) which resulted in the increase ofMDA (P〈0.05) with the increase of EE2 dosage. On the contrary, SOD, GSH-Px, CAT, T-AOC and MAD were unchanged (P〉0.05) in POP group. EE2 decreased antioxidase activity and closely related to the content of reactive oxygen species (ROS). And POP had the effect of anti-oxidative damage, protected the balance of ROS and the defense system ofantioxidant enzyme.
