摘  要:目的观察退变腰椎间盘髓核组织中转化生长因子-α(TGF-α)、基质金属蛋白酶-9(MMP-9)的表达并探讨其意义。方法标本分别取自退变组和对照组。所有标本一份制作为石蜡切片,用免疫组织化学方法检测TGF-α、MMP-9蛋白表达情况;另一份用RT-PCR方法检测TGF-α、MMP-9mRNA表达情况。结果退变组腰椎间盘髓核组织中TGF-α、MMP-9的表达量均较对照组增多(蛋白水平:t=29.114,P<0.01;t=19.926,P<0.01。mRNA水平:t=47.642,P<0.01;t=26.987,P<0.01),TGF-α和MMP-9蛋白、mRNA表达均呈正相关关系(蛋白水平:r=0.725,P<0.01;mRNA水平:r=0.684,P<0.01)。结论 TGF-α、MMP-9在正常与退变椎间盘髓核组织中表达量不同,退变椎间盘中二者表达量较高,两者可能与椎间盘退变有关。TGF-α对椎间盘退变的调节作用与MMP-9有一定的关系。Objective To investigate the 6xpression and its significance of TGF-a and MMP-9 in human nucleus pulposus of the degenerated human intervertebral discs. Methods The simples were from the experimental group and the control group. One part was made as paraffin section, using IHC method to measure the protein expression of TGF-a and MMP-9 The other part was using RT-PCR to measure the expression of TGF-a and MMP-9 mRNA. Results The results of RT-PCR and immu- nohistochemical staining showed that there were stronger expressions of TGF-a and MMP-9 in experimental group than those in control group (protein level: t=29.114, P〈0.01; t=19.926, P^0.01. mRNA level: t=47.642, P〈0.01; t=26.987, P〈0.01). There was certain positive correlation between the mRNA and protein expressions of TGF-a and those of MMP-9 in experimental group (protein level: r=0. 725, P〈0. 01. mRNA level: r=0. 684, P〈0.01). Conclusion There is significant difference in the expression level of TGF-a and MMP-9 between normal and degenerated lumbar disc. The effect of TGF-a on de- generated vertebral disc is correlate to MMP-9.
