摘  要:目的观察活性氧(reactive oxygen species,ROS)在血府逐瘀汤影响人脐静脉内皮细胞(human umbilical vein endothelial cell,HUVEC)迁移中的作用。方法运用血清药理学方法,采用Boyden小室法检测血府逐瘀汤含药血清对HUVEC迁移的影响;运用ROS清除剂预处理细胞,分析ROS在血府逐瘀汤调节内皮细胞迁移能力中的作用;并分析药物对内皮细胞H2O2产生的影响。结果血府逐瘀汤含药血清能增强HUVEC细胞的迁移能力,促进其H2O2的生成;ROS的清除可抑制血府逐瘀汤含药血清对细胞迁移的促进作用。结论血府逐瘀汤可通过激活ROS,增强HUVEC细胞的迁移能力,从而发挥促血管新生作用。Objective To investigate the role of reactive oxygen species (ROS) in the human umbilical vein endothelial cell (HUVEC) migration induced by Xuefu Zhuyu Decoction (XFZYD). Methods The effects of XFZYD treatment on the induction of the migration of HUVECs, as well as ROS scavenging on the XFZYD - induced cell migration, were observed by using hoyden chamber. Moreover, the effect of XFZYD on the H2 02 release from HUVECs was also assessed. Results Compared with the control group,XFZYD could significantly induce the migration of HUVECs and increase the endothelium - derived H2 02 concentration. Furthermore, the cell migration induced by XFZYD was inhibited by ROS scavenging, as showed in the experiment performed with ROS scavengers. Conclusion ROS plays an important role in the regulation of HUVECs migration induced by XFZYD.
