摘 要:目的探索披针新月蕨总黄烷对3T3-L1细胞增殖与分化的影响。方法通过检测细胞存活率来评价总黄烷的细胞毒性,通过检测油红O染色程度来评价总黄烷对3T3-L1细胞增殖分化的影响,通过实时定量逆转录多聚链反应(RT-PCR)检测细胞分化相关基因CCAAT增强子结合蛋白α(C/EBPα),脂肪型脂肪酸结合蛋白(FABP4)和葡萄糖转运因子4(GLUT-4)mRNA的表达情况。结果披针新月蕨给药组的细胞MTT值未有明显改变,而油红O染色程度显著性下降了,同时披针新月蕨给药明显的抑制了细胞C/EBPα,FABP4和GLUT-4基因的表达。结论披针新月蕨具有抑制3T3-L1细胞增殖与分化的活性。Objective The aim of this study was to investigate the effect of Abacopteris penangiana on the proliferation and differentiation of 3T3-L1 preadipocytes.Methods The cytotoxicity of Abacopteris penangiana was detected by MTT method.The proliferation and differentiation of preadipocytes were measured by oil and red O staining.Results No cytotoxicity was observed.The content of oil and red O staining of Abacopteris penangiana treated groups were significantly decreased.Furthermore,Abacopteris penangiana also inhibited the mRNA expressions of CCAAT/enhancer-binding protein α(C/EBPα),fatty acid binding protein(FABP4) and glucose transporter 4(GLUT4).Conclusion Abacopteris penangiana had the activity of inhibiting the proliferation and differentiation of 3T3-L1 preadipocytes.
